摘要
目的评价断血流提取物的止血作用,为进一步开发其外用止血产品提供依据。方法采用加热回流法制备断血流提取物,新西兰兔心脏取血,离心取上清液备用,实验分为3组:生理盐水组、断血流提取物组和云南白药组,通过测定血小板黏附率和血浆复钙时间来评价断血流的体外止血作用。将大鼠和新西兰兔各分为3组:空白对照组、断血流浸膏组和云南白药粉末组,建立大鼠股静脉出血、兔耳动脉出血模型,记录出血量和止血时间来探究断血流提取物的体内止血作用。结果与生理盐水组相比,断血流提取物组和云南白药组的血小板黏附率明显提高(P<0.05),血浆复钙时间明显缩短(P<0.05),但是断血流提取物组和云南白药组间差异无统计学意义(P>0.05)。在大鼠股静脉止血实验和新西兰兔耳动脉止血实验中,与空白对照组相比,断血流浸膏组和云南白药粉末组止血时间明显减少(P<0.05),但出血量没有明显差异。结论通过对断血流进行体内外止血效果评价,表明断血流具备优异的止血性能。
Objective To explore the hemostatic effect of the extract from Chinese Clinopodium herb,and provide a basis for the further development of its external hemostatic products.Methods The extract of Chinese Clinopodium herb was prepared by heating reflux method,and the supernatant was prepared by collecting blood from the heart of New Zealand rabbits and centrifuging at 1000 r/min.The experiment was randomly divided into three groups:normal saline group,extract of Chinese Clinopodium herb group and Yunnan Baiyao group,and the platelet adhesion rate and the plasma recalcification time were measured to evaluate the hemostatic effect in vitro of Chinese Clinopodium herb.Rats and New Zealand rabbits were randomly divided into three groups:blank control group,extract of Chinese Clinopodium herb group and Yunnan Baiyao powder group,and rat femoral vein hemorrhage model and rabbit ear artery hemorrhage model were established to measure the bleeding volume and the hemostasis time for the hemostatic effect in vivo of the extract,respectively.Results In the evaluation experiment of hemostasis in vitro,the platelet adhesion rate in extract of Chinese Clinopodium herb group and Yunnan Baiyao group was significantly higher than that in normal saline group(P<0.05),while the plasma recalcification time was significantly shorter(P<0.05).But there was no significant difference in platelet adhesion rate and plasma recalcification time between extract of Chinese Clinopodium herb group and Yunnan Baiyao group(P>0.05).In the hemostasis experiments of femoral vein in rats and auricular artery in New Zealand rabbits,compared with blank control group,the hemostasis time in extract of Chinese Clinopodium herb group and Yunnan Baiyao group was significantly shortened(P<0.05),but there was similar in the bleeding volume.Conclusion The results suggest that the Chinese Clinopodium herb has excellent hemostatic performance.
