Quality evaluation of Poria cocos in five producing areas

摘要 目的考察安徽、湖北和云南3个地区5个产地茯苓药材和饮片质量,为我国茯苓主产区药材质量研究和后续产业开发提供实验研究基础。方法首先采用性状鉴别法对云南腾冲、安徽金寨、湖北罗田、安徽霍山、安徽岳西5个产地的茯苓进行性状描述,采用显微鉴别和理化鉴别法分别展现其结构和理化特征,然后采用薄层色谱法对茯苓进行定性鉴别,最后利用高效液相色谱法测定其中茯苓酸的含量。结果在性状方面,5个产地茯苓药材性状与药典描述一致。5个产地的茯苓均具有药典规定的特征显微结构,理化鉴定均显深红色,其中安徽霍山茯苓粉末中不规则团块较多,理化显色也更明显。在薄层色谱鉴别方面,5个产地茯苓与相应对照药材色谱在相同位置上显示相同颜色主斑点。在含量方面,安徽霍山的茯苓中茯苓酸的含量在5个产地中最高,平均含量约为其他四地的1.05~1.68倍。结论通过对5个产地茯苓的鉴别和质量评价,为茯苓药材和饮片的质量标准建立、产地选择、新产品开发提供参考依据。 Objective To investigate the quality of Poria cocos and its medicine materical crude slices from 5 producing areas in Anhui,Hubei and Yunnan,and provide a basis for the quality research of Poria cocos in the main producing areas of China and the subsequent industrialization development.Methods Firstly,the characters of Poria cocos from Tengchong in Yunnan Province,Jinzhai in Anhui Province,Luotian in Hubei Province,Huoshan in Anhui Province and Yuexi in Anhui Province were described by character identification.The structure and physical and chemical characteristics of all Poria cocos were revealed by microscopic identification and physical-chemical identification,respectively.Then,the qualitative identification of Poria cocos was determined by thin layer chromatography(TLC).Finally,the content of pachymic acid was determined by high performance liquid chromatography(HPLC).Results The characters of Poria cocos from five producing areas were consistent with those described by Chinese Pharmacopoeia.All Poria cocos had the microstructures stipulated by Chinese Pharmacopoeia,and all were deep red color.Among them,the powder of Poria cocos form Huoshan in Anhui had more irregular clumps and showed more obvious physicochemical color.In the TLC identification,the main spots of the same color were found in the same position of Poria cocos from the five origins and the corresponding control herbs.In the five producing areas,the content of pachymic acid in Huoshan was the highest,and the average content in Huoshan was about 1.05-1.68 times that in the other four producing areas.Conclusion This study on the identification and quality evaluation of Poria cocos from five producing areas will provide an experimental reference for the establishment of quality standards,selection of producing areas and development of new products of Poria cocos.
