摘要
旨在探讨谷胱甘肽硫转移酶K1(Glutathione S-Transferase Kappa1,GSTK1)基因序列特征.以牦牛为试验对象,通过RT-PCR技术扩增GSTK1基因序列并分析生物信息学,采用RT-qPCR技术构建其在牦牛不同组织中的表达谱.ORF分析显示,牦牛GSTK1基因编码区为618 bp,编码氨基酸为205个.系统发育树分析显示,牦牛与野牦牛(99.68%)、黄牛(99.68%)和野牛(99.40%)之间的同源性最高,与北极熊(87.63%)、野猪(87.25%)和马(87.01%)之间的同源性最低.蛋白预测分析显示,GSTK1蛋白为不稳定的亲水蛋白,存在12个磷酸化位点.高级结构预测分析显示,GSTK1蛋白的主要成分为α-螺旋(50.73%)、无规卷曲(40.00%)和延伸链(9.27%).组织表达分析发现GSTK1在所检测牦牛卵巢、小肠、脾、肝、肾、肌肉、肺和心组织中均有表达,其中表达量最高的组织是肝,表达量最低的组织是脾和心,且与其他组织差异显著(P<0.05).本研究获得了牦牛GSTK1基因的序列特征及组织表达谱,为进一步挖掘牦牛GSTK1基因的功能提供参考依据.
The aim was to explore the gene sequence characteristics of Glutathione S-Transferase Kappa1(GSTK1).Taking yak as the test object, the CDS region of GSTK1 gene was specifically amplified by RT-PCR(Reverse-transcription PCR) followed by bioinformatics analysis, and its expression profiles in different tissues of yaks were constructed by RT-qPCR(Real Time Quantitative PCR) technique.The ORF analysis showed that the yak GSTK1 gene coding region was 618 bp, encoding 205 amino acids.Phylogenetic tree analysis showed that yak had the highest homology with wild yak(99.68%),cattle(99.68%) and bison(99.40%) and the lowest with polar bear(87.63%),wild boar(87.25%) and horse(87.01%).Protein prediction analysis showed that GSTK1 protein is an unstable hydrophilic protein with 12 potential phosphorylation sites.Advanced structure prediction analysis showed that the major components of GSTK1 protein were α-helix(50.73%),random coiled(40.00%) and strand(9.27%).Through tissue expression profile analysis, it was found that GSTK1 gene was expressed in 8 tissues of yak, including heart, liver, spleen, ovary, kidney, muscle, small intestine and lung, the tissue with the highest relative expression of GSTK1 was the liver, and the tissue with the lowest relative expression was the spleen and the heart, which varied significantly from other tissues(P<0.05).This study successfully obtained the sequence characteristics and tissue expression pattern of yak GSTK1 gene, which provided a reference for further exploring the function of yak GSTK1 gene.
