高强度紫外线对周丛生物膜除磷以及光合作用的影响研究

摘要 散养牲畜的排泄物是青藏高原局部地区水环境的重要污染源,排泄物中含有大量磷元素,流入水体可能引起水体富营养化,造成水华等,影响水生态环境以及牧民的生产生活.周丛生物膜对水体中磷等营养盐元素有良好的净化作用,但是,青藏高原地区强紫外线对周丛生物膜生长有何影响,进而是否会影响周丛生物对污染水体中磷元素的去除还鲜有报道.因此,本研究制备了牦牛粪便污水,模拟青藏高原地区UVA、UVB强度,研究其对周丛生物膜去除畜源性污水中不同形态磷的影响,以及UVA、UVB对周丛生物膜光合作用的影响.结果显示,暴露于高强度UVA、UVB下周丛生物膜对污水中的磷元素有较高的去除效率.暴露组总磷(TP)、总溶解性磷(TDP)、溶解性活性磷(SRP)和颗粒磷(PP)的平均去除效率高于70%,与控制组相比无显著差异.UVA和UVB照射不会阻碍周丛生物膜除磷功能.但是,UVA、UVB会抑制周从生物膜的光合作用,且UVB的抑制作用强于UVA,经过12 h UVA、UVB照射后,周丛生物膜的光合效率分别降低了20%和48%,而周丛生物膜可以通过自身修复抵抗高强度紫外线对其光合作用的损伤.本研究结果可促进周丛生物膜运用于高紫外线地区的污水净化研究,为青藏高原地区畜源性污水净化提供数据和理论支持. The excreta of free range livestock is an important pollution source of the water environment in some areas of the Qinghai Tibet Plateau.The excreta contains a large amount of phosphorus, which may cause eutrophication of the water body and cause water blooms, affecting the water ecological environment, the production and life of herdsmen.However, there are few reports on the impact of strong ultraviolet radiation on the growth of periphyton in the Qinghai Tibet Plateau.Therefore, yak fecal sewage was prepared in this study, treated by UV simulating the intensity of UVA and UVB in the Qinghai Tibet Plateau, and assayed its impact on the removal of different forms of phosphorus in livestock derived sewage by periphyton, and the impact of UVA and UVB on the photosynthesis of periphyton.The results showed that the periphyton exposed to high intensity UVA and UVB had high removal efficiency of phosphorus in sewage.The average removal efficiency of total phosphorus(TP),total dissolved phosphorus(TDP),soluble active phosphorus(SRP) and particulate phosphorus(PP) in the exposure group was higher than 70%,and there was no significant difference compared with the control group.UVA and UVB irradiation would not hinder phosphorus removal function from the periphyton.However, UVA and UVB could inhibit the photosynthesis of periphyton, and the inhibition effect of UVB was stronger than UVA.After 12 hours of UVA and UVB irradiation, the photosynthetic efficiency of periphyton decreased by 20% and 48% respectively, while periphyton could resist the photosynthesis damage caused by high intensity ultraviolet light through self-repair.The results of this study can promote the application of periphyton in the study of sewage purification in high UV areas, and provide data and theoretical support for the purification of livestock derived sewage in the Qinghai Tibet Plateau.
