摘要
以热带海草床沉积物土著硫氧化微生物资源的开发为目的,首先选择培养基并对海南文昌海草床沉积物中可培养硫氧化细菌进行了富集培养和分离纯化,进而利用16S rRNA测序方法获得了硫氧化细菌的分类信息并建立了系统发育树,最后对其硫氧化性质进行检测,共分离鉴定出38株具有硫氧化功能的菌株,它们均来源于变形菌门(Proteobacteria)中的γ-变形菌纲(Gamma-Proteobacteria)和厚壁菌门(Firmicutes).其中弧菌属(Vibrio)、芽孢杆菌属(Bacillus)和产微球茎菌属(Microbulbifer)为优势属,同时获得了嗜盐单胞菌属(Halomonas)、海杆菌属(Marinobacter)和深海海旋菌属(Thalassospira)中常见的硫化物氧化细菌.测序分析结果表明,海南文昌海草床环境样品中硫氧化细菌的多样性丰富,且不同站位之间受沉积物间隙水硫化物浓度的影响也存在差异.本研究获得了海南文昌海草床不同类型的硫氧化细菌菌株,丰富了海南海草床土著硫氧化细菌的菌种资源,为海草床沉积物硫化物微生物治理技术的开发奠定了基础.
In the report,aimed at exploiting the indigenous sulfur-oxidizing microbial strains in tropical seagrass meadows sediments,the medium was screened,and the culturable sulfur-oxidizing bacteria were enriched,isolated and purified from the sediments of seagrasses meadow in Wenchang,Hainan.16S rRNA sequencing was used to obtain the classification information of these sulfur oxidizing bacteria strains and a phylogenetic tree was constructed.The sulfur oxidation properties of these strains were tested.A total of 38 sulfur oxidizing strains were isolated and identified,and all of them were belonged toγ-Proteobacteria and Firmicutes.The Vibrio,Bacillus and Microbulbifer are the dominant genus.The sulfide-oxidizing bacteria commonly found in Halomonas,Marinobacter and Thalassospira.were obtained.Sequencing analysis result indicated that the sulfur oxidizing bacteria were abundant in sediments of Wenchang seagrasses meadow in Hainan.Our study obtained sulfur oxidizing bacteria of different types from seagrass meadow in Wenchang,which will lay a foundation for the development of sulfide microbial treatment technology for ecological remediation seagrass meadow sediments.
