响应面法优化虎尾轮根中总黄酮提取工艺

摘要 采用响应面法优化虎尾轮根中总黄酮的提取工艺。综合比较各种提取方法,选择超声波-微波提取法,在单因素实验基础上,确定超声波-微波反应时间、乙醇体积分数、提取温度为响应因素,以虎尾轮总黄酮提取率为响应值,按照BoxBenhnken试验设计采用三因素三水平方法完成响应面试验,观察各因素对总黄酮提取率的影响。各因素对虎尾轮根中总黄酮提取的影响顺序为提取温度>乙醇体积分数>提取时间。响应面优化的最佳提取工艺:超声波-微波提取时间6 min,乙醇体积分数72%,反应温度72℃,虎尾轮中总黄酮提取率为1.635%。平行实验3次,实际得到的虎尾轮根中总黄酮提取率为1.625%、1.634%、1.647%,平均提取率为1.635%,相对标准偏差为0.9573%。改进后的提取工艺比传统的提取方法操作更简单,且稳定高效。 Response surface methodology(RSM)was used to optimize the extraction process of total flavonoids from the root of crinite uraria.By comprehensively comparing various extraction methods,ultrasonic-microwave extraction method was selected,on the basis of single factor experiment,ultrasonic-microwave reaction time,ethanol volume fraction and extraction temperature were determined as the response factors,and the extraction rate of total flavonoids was taken as the response value.According to Box-Benhnken experiment design,three-factor and three-level method was used to complete the response surface experiment,and the effects of each factor on the extraction rate of total flavonoids were observed.The order of the effects of various factors on the extraction of total flavonoids from the root of crinite uraria was extraction temperature>ethanol volume fraction>extraction time.The optimal extraction conditions were as follows:ultrasonic-microwave extraction time was 6 min,ethanol volume fraction was 72%,reaction temperature was 72℃,and the total flavonoids extraction rate was 1.635%.After three parallel experiments,the actual extraction rates of total flavonoids from the root of crinite uraria were 1.625%,1.634%and 1.647%,the average extraction rate was 1.635%,and the relative standard deviation was 0.9573%.The improved extraction process is more simple and stable than traditional extraction method.
