摘要
目的:基于网络药理学的研究思路及方法探索中药金银花治疗痤疮的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)筛选金银花活性成分及对应靶点,利用GeneCards、OMIM、TTD数据库进行检索得到与痤疮有关的靶点基因,通过Venny 2.1.0得到药物与疾病的共同靶点。借用Cytoscape 3.9.0绘制金银花活性成分-相关靶点网络图,采用STRING数据库构建蛋白质-蛋白质相互作用网络,将药物-疾病共同靶点通过Metascape数据库分别进行基因本体论(GO)富集分析及京都基因与基因组百科全书(KEGG)通路富集分析。结果:本研究共筛选得出槲皮素、木犀草素、豆甾醇、山柰酚以及β-谷甾醇等23个活性成分,药物靶点211个,痤疮相关靶基因612个,主要涉及主要涉及肿瘤坏死因子(Tumor Necrosis Factor,TNF)、白蛋白(Albumin,ALB)、白细胞介素(Interleukin,IL)-6、肿瘤蛋白p53(Tumor Protein p53,TP53)、IL-1β、基质金属蛋白酶9(Matrix Metallopeptidase 9,MMP9)等43个关键靶点,通过丝裂原活化蛋白激酶(Mitogen-activated Protein Kinase,MAPK)信号通路、磷脂酰肌醇-3-激酶(Phosphatidylinositol 3-kinase,PI3K)-丝氨酸/苏氨酸蛋白激酶(Akt Serine/threonine Protein Kinase,Akt)信号通路、乙型肝炎等主要信号通路发挥治疗作用。结论:本研究基于网络药理学的原理初步探讨了金银花治疗痤疮的核心靶点与信号通路,金银花通过多种成分、多个靶点、多条通路实现干预痤疮治疗的作用。旨在为此后金银花在痤疮中的临床应用提供参考和指导。
Objective:Based on the research ideas and methods of network pharmacology,to explore the mechanism of honeysuckle in the treatment of acne.Methods:The active components and corresponding targets of Jinyinhua(Flos Lonicerae Japonicae) were screened through TCMSP database,and the target genes related to acne were retrieved by Gene Cards,OMIM and TTD database.The common targets of drugs and diseases were obtained through Venny 2.1.0.Cy to scape 3.9.0 software was used to draw the network diagram of the active components of Jinyinhua and the related targets,the protein-protein interaction network was constructed by STRING database,and the drug-disease common targets were enriched and analyzed by GO and KEGG pathways through Metascape database.Results:In this study,23 active components including quercetin luteolin,stigmasterol,kaempferol and b-sitosterol were screened,including 211 drug targets and 612 acne related target genes,mainly involving 43 key targets such as TNF,ALB,IL-6,TP53,IL-1β and MMP9.They play a therapeutic role through MAPK signal pathway,PI3K-Akt signal pathway,hepatitis B signal pathway and so on.Conclusion:Based on the principle of network pharmacology,this study preliminarily discussed the core targets and signal pathways of Jinyinhua in the treatment of acne.Jinyinhua can intervene the treatment of acne through multiple components,multiple targets and multiple pathways.This study provides more reference and guidance for the clinical application of Jinyinhua in acne.
