重楼正丁醇部位指纹图谱与体外抗氧化作用的谱-效关系研究 被引量:1

Study on the Fingerprint of n-Butanol Fraction of Paridis Rhizoma and Its Anti-oxidant Activity Spectrum-Effect Relationship

摘要 目的建立重楼正丁醇部位的UPLC指纹图谱,考察其化学成分与抗氧化活性的谱-效关系。方法采用UPLC法建立12批重楼正丁醇部位的指纹图谱,并进行共有峰指认及相似度评价;通过DPPH法和ABTS法测定其抗氧化活性,计算IC50值;将12批重楼正丁醇部位共有峰数据与抗氧化活性进行皮尔逊相关分析和逐步回归分析,得到二者的谱-效关系。结果建立了12批重楼正丁醇部位UPLC指纹图谱,共确定了14个共有峰,相似度均大于等于0.958;采用对照品比对法指认了6号峰为重楼皂苷Ⅶ,7号峰为重楼皂苷Ⅵ,11号峰为重楼皂苷Ⅱ,12号峰为重楼皂苷Ⅰ。结论本研究成功建立了重楼正丁醇部位的UPLC指纹图谱,明确了重楼正丁醇部位的体外抗氧化活性,重楼皂苷Ⅰ、重楼皂苷Ⅱ、重楼皂苷Ⅶ对抗氧化活性贡献度较大,可能是其抗氧化活性的主要活性成分。 Objective To establish the UPLC fingerprint of n-butanol fraction of Paridis Rhizoma,and investigate the spectral effect relationship between n-butanol fraction and anti-oxidant activity.Methods UPLC method was used to establish the fingerprint of n-butanol in 12 batches of Paridis Rhizoma,and the common peak identification and similarity evaluation degree were carried out.DPPH and ABTS methods were used to determine the anti-oxidant activity and the IC50 value was calculated.Pearson correlation analysis and stepped-regression analysis were performed on the common peak data and anti-oxidant activity of n-butanol fractions in 12 batches.Results 12 batches of UPLC fingerprints of n-butanol extraction parts were established,and 14 common peaks were determined with similarity≥0.958.PolyphyllinⅦ(Peak 6),PolyphyllinⅥ(peak 7),PolyphyllinⅡ(peak 11)and Polyphyllin I(peak 12)were identified by comparison method.Conclusion The UPLC fingerprint of n-butanol extract of Paridis Rhizoma is successfully established,and the anti-oxidant activity of it is confirmed.PolyphyllinⅠ,PolyphyllinⅡand PolyphyllinⅦare contributed significantly to the anti-oxidant activity,which may be the main components of its anti-oxidant activity.
