摘要
以绿萝叶片为试验材料,采用80%丙酮研磨法、96%乙醇和DMSO浸提法提取叶绿素,用紫外分光光度法对叶绿素含量进行测定,研究提取方法对叶绿素提取量的影响。结果发现:三种方法得到的叶绿素提取液的紫外光谱相似,在665-667nm有最大吸收峰值,但是吸收强度不同。80%丙酮研磨法所得叶绿素含量最低,96%乙醇和DMSO浸提法所得叶绿素含量较高,但差异不显著。表明96%乙醇和DMSO是良好的叶绿素提取溶剂,但用96%乙醇浸提法所得叶绿素提取液稳定性稍差,要注意避光保存。比较不同方法的测定结果,分析其优缺点,为今后研究选择合适的叶绿素提取方法提供依据。
Adopting the leaves of Epipremnum aureum as the test material,and using the 80%acetone grinding method and 96%ethanol and DMSO extraction method respectively to measure the chlorophyll level,the paper puts forward the following findings by comparing the results with different measuring methods:the absorption spectra of the chlorophyll extract obtained by the three methods are similar;the 80%acetone grinding method leads to lower measuring values,while there is no significant difference between the 96%ethanol method and the DMSO extraction method.Both 96%ethanol and DMSO are good solvents for chlorophyll extraction,but the chlorophyll extract obtained by the 96%ethanol extraction method has poor stability,so exposure to light should be avoided.
