用高通量测序技术分析红皮云杉梢枯病前后叶围微生物多样性 被引量:1

High-throughput Sequencing Analysis of Microbial Diversity before and after Infection with Spruce Dieback

摘要 采用Illumina Miseq高通量测序技术来探究红皮云杉(Picea koraiensis)梢枯病(Dothiorella gregaria)病叶与健康叶之间共生真菌和细菌群落结构的差异。结果表明:红皮云杉叶片感病前后细菌的OTU变化巨大,真菌的OTU变化也偏大。两组样品共有的细菌种类45个,占细菌总数的16.13%,真菌种类73个,占真菌总数的29.32%。两组样品中菌落结构基本相同,但相对丰度差异较大,在门水平下,细菌中变形菌门(Proteobacteria)在两组样品占据绝对优势,真菌中子囊菌门(Ascomycota)在两组样品占据绝对优势。在属水平下,克雷伯菌属(Klebsiella)是健康叶中的优势细菌,马赛菌属(Massilia)在病叶中占据绝对优势;亚隔孢壳属(Didymella)是健康叶及病叶中的优势真菌,在健康叶及病叶中优势真菌种类未有太大变化,小穴壳菌属(Dothiorella)及Naganishia属仅存在于病叶组。 Illumina Miseq high-through Out sequencing technology was used to explore the differences in the community structure of symbiotic bacteria and fungi between diseased and healthy leaves of Picea koraiensis(Dothiorella gregaria).The OTU of bacteria changed greatly before and after infection of spruce leaves,and the OTU of fungi also changed greatly.There were 45 bacterial species in the two groups of samples,accounting for only 16.13%of the total number of bacteria,and 73 fungal species in total,accounting for only 29.32%of the total number of fungi.The colony structures of the two groups of samples were basically the same,but the relative abundances were quite different.At the phylum level,in bacteria Proteobacteria dominated the two groups of samples,and in fungi,Ascomycota dominated the two groups of samples.At the genus level,Klebsiella is the dominant bacteria in healthy leaves,Massilia is dominant in diseased leaves;Didymella is the dominant fungi in healthy and diseased leaves.There was no significant change in the species of dominant fungi in healthy and diseased leaves.Only Dothiorella and Naganishia were found in the diseased leaf group.
