GC法同时测定天麻追风膏中3种挥发性成分的含量

摘要 目的:建立气相色谱法同时测定天麻追风膏中龙脑、异龙脑和樟脑含量的方法。方法:以聚乙二醇20000(PEG-20M)为固定相的弹性石英毛细管柱岛津SH-Rtx-Wax(30 m×0.32 mm×0.25μm),载气为高纯氮气(≥99.999%),FID检测器。进样口温度220℃,检测器温度250℃,分流比15∶1。程序升温:初始温度80℃,保持1 min,以每分钟20℃的速率升温至120℃,保持1 min,以每分钟5℃的速率升温至160℃,保持8 min。进样为1μL。结果:龙脑、异龙脑和樟脑分别在7.828~1002,7.758~993.0,3.944~504.9μg·mL^(-1)内呈良好的线性关系;平均加样回收率分别为98.3%(n=6,RSD=1.4%);95.9%(n=6,RSD=1.5%);102.8%(n=6,RSD=1.0%)。测定样品12批,结果龙脑、异龙脑和樟脑的含量分别在3.69~266.8、2.6~151.5和8.89~240.6μg·g^(-1)之间。结论:所建立的方法可用于天麻追风膏中冰片的质量控制,且为完善天麻追风膏的质量标准提供了科学依据。 Objective:To establish a method for simultaneous determination of borneol,isoborneol and camphor in Tianma Zhuifeng ointment by gas chromatography.Methods:Shimadu SH-Rtx-Wax(30 m×0.32 mm×0.25μm)with polyethylene glycol 20000(PEG-20M)as stationary phase,high purity nitrogen(≥99.999%)as carrier gas,FID detector.The inlet temperature was 220℃,the detector temperature was 250℃,and the split ratio was 15∶1.Program heating:initial temperature 80℃,keep for 1 min,at the rate of 20℃per minute to 120℃,keep for 1 min,at the rate of 5℃per minute to 160℃,keep for 8 min.The injection sample was 1μL.Results:The linear relationship of borneol,isoborneol and camphor was 7.828-1002,7.758-993.0,3.944-504.9μg·mL^(-1),respectively;The average recoveries were 98.3%(n=6,RSD=1.4%);95.9%(n=6,RSD=1.5%);102.8%(n=6,RSD=1.0%).The contents of borneol,isoborneol and camphor in12 batches were 3.69-266.8,2.6-151.5 and 8.89-240.6μg·g^(-1),respectively.Conclusion:The established method can be used for the quality control of borneol in Tianma Zhuifeng ointment,and provide a scientific basis for improving the quality standard of Tianma Zhuifeng ointment.
