摘要
目的:研究核桃楸(Juglans mandshurica Maxim)树皮、枝皮和果皮的化学成分。方法:核桃楸皮90%乙醇提取物,分别用石油醚、二氯甲烷、正丁醇萃取,各萃取部位再采用反复硅胶柱色谱、反相ODS柱色谱、半制备HPLC及重结晶等手段进行分离;结合ESI-MS和1D,2D-NMR波谱数据鉴定化合物结构。结果:分离得到12个化合物,分别鉴定为(4S)-4-羟基-α-四氢萘酮-4-O-β-D-(6′-O-4″-羟基苯甲酰)吡喃葡萄糖苷(1)、(4S)-4,5-二羟基-α-四氢萘酮-4-O-β-D-(6′-O-4″-羟基苯甲酰)吡喃葡萄糖苷(2)、(4S)-4,5,8-三羟基-α-四氢萘酮-5-O-β-D-(6′-O-4″-羟基苯甲酰)吡喃葡萄糖苷(3)、(4R)-4-羟基-5-甲氧基-α-四氢萘酮-4-O-β-D-吡喃葡萄糖苷(4)、(4S)-4,5,8-三羟基-α-四氢萘酮-5-O-(6′-O-α-L-阿拉伯糖基)-β-D-吡喃葡萄糖苷(5)、4(S)-4,5,8-三羟基-α-四氢萘酮-5-O-(6′-β-D-吡喃葡萄糖基)-β-D-吡喃葡萄糖苷(6)、胡桃醌(7)、5,8-二羟基-1,4-萘醌(8)、核桃酮(9)、没食子酸(10)、香草酸(11)和丁香酸(12)。结论:化合物1~6均为首次从核桃楸皮药用部位中分离得到。
Objective:To study the chemical constituents in barks of Juglans mandshurica Maxim.Methods:90%ethanol extracts from barks were extracted with petroleum ether,dichloromethane and n-butanol,respectively.The extracts were separated by repeated silica gel column chromatography,reversed-phase ODS column chromatography,semi-preparative HPLC and recrystallization.The structures of these compounds were determined by ESI-MS and 1D,2D-NMR spectral data analyses.Results:Twelve compounds were identified as(4S)-4-hydroxy-α-tetralone-4-O-β-D-(6′-O-4″-hydroxylbenzoyl)glucopyranoside(1),(4S)-4,5-dihydroxy-α-tetralone-4-O-β-D-(6′-O-4″-hydroxybenzoyl)glucopyranoside(2),(4S)-4,5,8-thihydroxy-α-tetralone-5-O-β-D-(6′-O-4″-hydroxylbenzoyl)glucopyranoside(3),(4R)-4-dihydroxy-5-methoxy-α-tetralone-4-O-β-D-glucopyranoside(4),(4S)-4,5,8-trihydroxy-α-tetralone-5-O-(6′-O-α-Larabinopyranosyl)-β-D-glucopyranoside(5),4(S)-4,5,8-thihydroxy-α-tetralone-5-O-(6′-O-β-D-glucopyranosyl)-β-D-glucopyranoside(6),juglone(7),5,8-dihydroxy-1,4-naphthoquinone(8),regiolone(9),gallic acid(10),vanillic acid(11)and syringic acid(12).Conclusion:Compounds 1-6 were isolated from barks of Juglans mandshurica Maxim for the first time.
