摘要
试验以9株产朊假丝酵母(Candida utilis)作为出发菌株,通过对碳源、氮源利用情况及蛋白含量进行综合分析,选择1株综合性能较好的菌株为出发菌株,进行C离子束辐照诱变和高蛋白含量突变菌株的筛选。结果显示:采用加权平均数的方法系统地对9株产朊假丝酵母进行评分,筛选出Candida utilis CGMCC2.2878为后续诱变育种的出发菌株。对其进行重离子诱变,通过筛选得到蛋白含量较高的突变菌株,命名为Y-4-2,其蛋白含量高达63.06%,较出发菌株(52.29%)提高了20.60%。研究表明,分离菌株有望应用于发酵生产菌体蛋白饲料。
In the experiment,nine strains of Candida utilis as the starting strains,through comprehensive analysis of carbon source,nitrogen source utilization and protein content,a strain with better comprehensive performance was selected as the starting strain forCion beam irradiation mutagenesis and screening of mutant strains with high protein content.The results showed that nine strains of Candida utilis were systematically scored by the method of weighted average,and Candida utilis CGMCC2.2878 was screened out as the starting strain for subsequent mutation breeding,which was subjected to heavy ion mutagenesis,the mutant strain with higher protein content was obtained by screening,named Y-4-2,and its protein content was as high as 63.06%,which was 20.60%higher than that of the starting strain of 52.29%.The study indicates that the isolated strain is expected to be used in fermentation production of bacterial protein feed.
