芦笋茎皮喷雾干燥工艺优化及化学成分的超高效液相色谱-串联质谱分析

Optimization of Spray Drying Process and Chemical Composition Analysis of Asparagus Stem Bark by UHPLC-Tandem Mass Spectrometry Method

摘要 芦笋茎皮是芦笋在生产加工过程中产生的废弃物,急需科学有效的方式实现芦笋废弃物的资源化利用。基于此,以绿芦笋茎皮为原料,考察进风温度、进样流量和料液密度,优化芦笋茎皮提取物喷雾干燥最优工艺;并利用超高效液相-四级杆-飞行时间高分辨质谱联用(UHPLC-Q-TOF-MS/MS)技术对喷雾干燥粉的化学成分进行分析。结果表明:芦笋茎皮提取物喷雾干燥的最优工艺为进风温度130℃、进样流量350 mL·h^(-1)、料液密度1.07 g·cm^(-3)。最优条件下平行制备的3批喷雾粉,其外观呈乳白色或淡黄色粉末,溶化性、润湿性、含水量、溶解度符合有关规定;芦笋茎皮喷雾粉中共鉴定到38个化合物,其中皂苷类化合物5个、黄酮类化合物8个、氨基酸8种、有机酸8个,以及其他类型化合物9个。综上所制备的芦笋茎皮喷雾粉可作为功能食品原料,提高了芦笋废弃资源利用率,促进了芦笋深加工产业发展。 Asparagus stem bark is the waste materials produced in the process of production and processing.It is urgent to study the resource utilization of asparagus waste in a scientific and effective way.To optimize the optimal spray drying process of asparagus stem bark extract by investigating the inlet air temperature,sample flow rate and liquid density using green asparagus stem bark as raw material;the spray dried powder was analyzed by ultra-high performance liquid chromatography-quadrupole-time-of-flight high-resolution mass spectrometry(UHPLC-Q-TOF-MS/MS).The results showed that the optimal spray drying parameters of asparagus stem bark was 130℃ for the inlet air temperature,350 mL·h^(-1) for the injection flow rate and 1.07 g·cm^(-3) for the solution density.Three batches of spray powders prepared in parallel under the optimal conditions showed the appearance of creamy white or light yellow,with the characteristic taste and odor of asparagus,and the dissolving property,wettability,water content and solubility were in accordance with the relevant regulations;a total of 38 compounds were identified in the spray powder of asparagus stem bark,including 5 saponins,8 flavonoids,8 amino acids,8 organic acids,and 9 other types of compounds.In conclusion,the spray drying powder prepared from the asparagus stem bark can be used as a functional food ingredient,which are helpful for improving the utilization of asparagus waste resources and promoting the industry development.
