香青兰乙醇提取物对溃疡性结肠炎大鼠AhR/I-κB相关通路蛋白及炎症因子表达的影响

Effects of Dracocephalum moldavica L.extract on the expression of AhR/I-κB related pathway proteins and inflammatory factors in rats with ulcerative colitis

摘要 目的:分析香青兰乙醇提取物对硫酸葡聚糖钠盐(DSS)诱导的溃疡性结肠炎(UC)芳香烃受体(AhR)、核转录因子-κb抑制蛋白(I-κB)通路蛋白及相关炎症因子表达的影响。方法:雄性SD大鼠30只,随机分为空白组、模型组和香青兰组,每组10只。除空白组外,其他各组大鼠饮用5%DSS溶液7 d后,香青兰组大鼠灌胃给予400 mg/kg香青兰乙醇提取物,模型组、空白组给予等量生理盐水灌胃,每天1次,连续给药7 d。记录小鼠体重、粪便性状及便血情况,HE染色观察结肠组织病理学变化。采用实时荧光定量PCR法检测AhR、IL-1β、IL-6和IL-8的mRNA水平,采用Western Blot检测结肠组织p-I-κB蛋白的表达。结果:与空白组比较,模型组大鼠体重减轻,出现黏液脓血便,结肠组织黏膜下水肿、出血、大量炎性细胞浸润,IL-1β、IL-6、IL-8表达升高(P<0.05),AhR表达降低(P<0.05),p-I-κB表达升高(P<0.05)。与模型组比较,香青兰组DAI评分降低,结肠病理改善,IL-1β、IL-6、IL-8表达明显降低(P<0.05),AhR表达升高(P<0.05),p-I-κB表达降低(P<0.05)。结论:香青兰乙醇提取物可调节AhR/I-κB信号通路,抑制促炎因子释放,减轻肠道炎症反应,从而对溃疡性结肠炎发挥保护作用。 Objective:To observe the effects of Dracocephalum moldavica L.(DML)extract on the expression of AhR,I-κB and related inflammatory factors in rats with ulcerative colitis.Methods:Total 30 male SD rats were divided into three groups(n=10/group)randomly as follows:control group,model group and DML group.Experimental colitis was induced in rats by oral administration of 5%DSS solution for 7 days.Beginning on Day 8,rats in DML group were given DML 400 mg/kg for 7 days.The control group and model group were given the same amount of physiological saline.The body weight,diarrhea degree and hematochezia were recorded daily.HE staining was used to observe the histopathological changes.The mRNA expression levels of AhR,IL-1β,IL-6 and IL-8 was detected by real time fluorescence quantitative PCR.The protein expression levels of p-I-κB in colon tissues were detected by Western blot.Results:Compared with control group,weight lost,had mucus,pus and blood in the stool,mucosal hemorrhage,inflammatory cell infiltration and submucosal edema were observed in model group.The expression levels of IL-1β,IL-6,IL-8,p-I-κB increased(P<0.05),while the expression level of AhR decreased(P<0.05).Compared with model group,the DAI score decreased,the pathological colon conditions were improved in DML group.The expression levels of IL-1β,IL-6,IL-8,p-I-κB decreased(P<0.05),while the expression level of AhR increased(P<0.05).Conclusion:Dracocephalum moldavica L.extract can regulate AhR and I-κB signaling pathways,inhibit the release of proinflammatory factors and reduce intestinal inflammatory response,so as to play a protective role in ulcerative colitis.
