摘要
花青素合成酶(anthocyanidin synthase,ANS)是植物花青素生物合成途径的关键酶,可催化无色花色素向有色花色素转变。根据已报道的花青素合成酶基因序列设计引物,分别以不同皮色洋葱苗期总DNA和cDNA为模板进行克隆和序列比对,结果显示,从紫皮和黄皮洋葱中克隆到相同的目的条带,基因序列完全一致,且与已报道的ANS等位基因序列高度相似,确认克隆到AcANS基因片段序列。以Actin基因为内参,对AcANS在两种皮色洋葱苗期不同组织中的表达进行半定量分析,结果表明,AcANS在洋葱幼苗叶鞘中表达量最高,叶片中次之,根中不表达;紫皮洋葱中AcANS基因的表达量明显高于黄皮洋葱,这有利于紫皮洋葱积累更多的花色苷,从而使鳞茎产生颜色上的差异。本研究结果可为后续获得洋葱ANS基因全长序列、深入研究其功能奠定基础。
Anthocyanidin synthase(ANS)is one of the key enzymes in plant anthocyanin biosynthesis pathway,which catalyzes the conversion of colorless anthocyanin to colored anthocyanin.We designed primers based on reported ANS gene sequences and used the total DNA and cDNA from seedlings of onion with different skin colors as templates for gene cloning and sequence alignment.The results showed that the same target band with identical sequence was cloned from the onion with purple and yellow skin,which was highly similar to the reported ANS allele sequences.Therefore,it could be confirmed that the AcANS gene sequence fragment was cloned.Using Actin gene as internal reference,the expression level of AcANS in different tissues of onion seedlings was analyzed semi-quantitatively.The results showed that the expression level of AcANS gene was the highest in leaf sheath,followed by leaves,and there was no expression in root.The expression level of AcANS gene in purple onion seedlings was significantly higher than that in yellow onion seedlings,which promoted the accumulation of anthocyanin and led to different color of bulbs.The results of this study could lay a foundation for obtaining the full length sequence of onion ANS gene and further studying its function.
