摘要
目的研究安肠汤低、中、高剂量对大鼠重症溃疡性结肠炎的抗炎作用,通过研究不同给药剂量对重症溃疡性结肠炎大鼠炎症因子水平的影响探讨安肠汤防治溃疡性结肠炎的可能性。方法实验大鼠分为空白组、模型组、美沙拉嗪组、丽珠肠乐组、安肠汤低剂量组、安肠汤中剂量组、安肠汤高剂量组,每组12只。除正常组外,其余各组均用三硝基苯磺酸(TNBS)/乙醇灌肠法建立溃疡性结肠炎大鼠模型,造模成功后,采集血清,结肠病变组织,PCR检测IL-10、IL-17、STAT3水平变化,蛋白免疫印迹法检测结肠组织中STAT3、磷酸化STAT3(p-STAT3)蛋白表达的变化,酶联免疫吸附测定检测各组大鼠血清IL-10、IL-17含量变化。结果与正常组比较,模型组大鼠STAT3信号通路中p-STAT3、STAT3蛋白表达显著增高(P<0.05),炎症相关因子如IL-10、IL-17水平升高(P<0.05),经过安肠汤干预的大鼠p-STAT3、STAT3蛋白以及IL-10、IL-17表达水平显著降低且与安肠汤的给药浓度有关(P<0.05)。结论安肠汤可通过影响STAT3信号转导通路相关基因、蛋白和炎症因子的表达,抑制溃疡性结肠炎病情的发展。
Objective To study the anti-inflammatory effects of Anchang Decoction in low,middle and high doses on severe ulcerative colitis in rats,and to explore the possibility of Anchang Decoction in preventing and treating ulcerative colitis by studying the effects of different doses on inflammatory factors in rats with severe ulcerative colitis.Methods The experimental rats were divided into blank group,model group,mesalazine group,Lizhu Changle group,Anchang Decoction low dose group,Anchang Decoction middle dose group and Anchang Decoction high dose group with 12 rats in each group.Except for the normal group,the rats of ulcerative colitis were established by trinitrobenzene sulfonic acid(TNBS)/ethanol enema in other groups.After the model was successfully established,serum and pathological tissues of colon were collected,and the levels of IL-10,IL-17 and STAT3 were detected by PCR.The expression of STAT3 and phosphorylated STAT3(p-STAT3)in colon tissues was detected by Western blot,and the serum IL of rats in each group was detected by enzyme-linked immunosorbent assay.Results The results of this study confirmed that compared with the normal group,the expression of p-STAT3 and STAT3 protein in STAT3 signaling pathway of rats in model group was significantly higher(P<0.05),and the levels of inflammatory factors such as IL-10 and IL-17 were higher(P<0.05).The p-STAT3 and STAT3 protein,IL-10 and IL-17 in rats treated with Anchang Decoction were significantly higher.(P<0.05).Conclusion Anchang Decoction can inhibit the development of ulcerative colitis by affecting the expression of STAT3 signal transduction pathway related genes,proteins and inflammatory factors.
