荔枝核总黄酮通过JAK2/STAT3信号通路抗大鼠肝纤维化的实验研究 被引量:7

Effects of total flavone from Litchi chinensis sonn on rats with hepatic fibrosis through JAK2/STAT3 signaling pathway

摘要 目的探讨荔枝核总黄酮(TFL)通过酪氨酸蛋白激酶2(JAK2)/信号转导及转录激活因子3(STAT3)信号通路对二甲基亚硝胺(DMN)诱导的大鼠肝纤维化的作用机制。方法将30只大鼠近照随机数字表法分为空白对照组、模型组、TFL组,各10只。除空白对照组外,其余各组大鼠采用DMN腹腔注射4周建立大鼠肝纤维化模型。造模后,TFL组给予TFL灌胃,空白对照组和模型组给予等体积生理盐水灌胃,6周后处死大鼠。检测大鼠血清天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)水平及肝组织羟脯氨酸(HYP)水平。取固定部位肝组织进行病理组织学检查,并采用Western blot检测JAK2、磷酸化JAK2(p-JAK2)、STAT3、磷酸化STAT3(p-STAT3)、α-平滑肌肌动蛋白(α-SMA)、胶原-Ⅰ(Collagen-Ⅰ)蛋白表达情况。结果与模型组比较,TFL组大鼠DMN诱导的肝损伤病理表现改善,血清AST、ALT及肝组织HYP水平降低(P<0.05)。Western blot检测结果显示,TFL组α-SMA、Collagen-Ⅰ、p-JAK2、JAK2、p-STAT3、STAT3蛋白表达较模型组降低(P<0.05)。结论TFL对DMN诱导的肝纤维化大鼠具有保护作用,其机制可能与JAK2/STAT3信号通路有关。 Objective To observe the effects of total flavone from Litchi chinensis sonn(TFL)on dimethylnitrosamine(DMN)-induced hepatic fibrosis in rats through janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods Thirty rats were divided into blank control group,model group and TFL group according to the random number table method,with 10 rats in each group.Except for the blank control group,the rats in other groups were intraperitoneally injected with DMN for 4 weeks to establish the hepatic fibrosis model.After establishment of the model,TFL group was given TFL by gavage,while blank control group and model group were given the same volume of normal saline by gavage for 6 weeks.The serum levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT)and hydroxyproline(HYP)of liver tissues were detected.Meanwhile,the liver tissue were collected for histopathological examination.The protein expressions levels of JAK2,phosphorylated JAK2(p-JAK2),STAT3,phosphorylated STAT3(p-STAT3),α-smooth muscle actin(α-SMA)and Collagen-Ⅰwere detected by Western blot.Results Compared with the model group,the pathological manifestations of DMN-induced liver injury of rats in TFL group were improved,and the levels of serum AST,ALT and HYP of liver tissue were decreased(P<0.05).The results of Western blot showed that p-JAK2,JAK2,p-STAT3 and STAT3,α-SMA,Collagen-Ⅰexpression levels in TFL group were decreased compared with those in model group(P<0.05).Conclusion TFL has a protective effect on DMN-induced hepatic fibrosis in rats,and its mechanism may be related to JAK2/STAT3 signaling pathway.
