不同来源金黄色葡萄球菌分离株溶血素基因分布分析

Analysis on distribution of hemolysin genes of Staphylococcus aureus from different sources

摘要 目的了解广西壮族自治区南宁市不同来源金黄色葡萄球菌分离株溶血素基因的分型情况和分布特点。方法收集南宁市辖区所有食品安全监测点分离的金黄色葡萄球菌216株,采用多重PCR扩增法对菌株进行hlα、hlb、hlg和hld四种溶血素基因检测及溶血试验。结果食源性分离株溶血素基因hlα、hlb、hlg和hld基因检出率分别为55.9%、72.1%、75.7%和83.8%,以携带hlb/hlg/hld基因组合为主,检出率为33.1%;从业人员携带者分离株溶血素基因hlα、hlb、hlg和hld基因检出率分别为78.8%、51.3%、87.5%和85.0%,携带以hlα/hlg/hld基因组合为主,检出率为37.5%。来源乳制品分离株hlb基因检出率高于米面制品分离株(P<0.01);餐饮从业人员携带者分离株hlα基因检出率高于食品企业生产人员携带者分离株(P<0.05);从业人员携带者分离株完全溶血检出率高于食源性分离株(P<0.05)。结论广西壮族自治区南宁市不同样本来源的金黄色葡萄球菌分离株溶血素基因检出率和分布模式存在差异,应加强溶血素基因监测体系建设。 Objective To understand the hemolysin gene distribution and classification of Staphylococcus aureus from different sources in Nanning city,Guangxi Zhuang autonomous region.Methods A total of 216 strains of Staphylococcus aureus isolated from all food safety monitoring sites in Nanning city were collected.The multiplex polymerase chain reaction was used to detect Staphylococcus aureus hemolysin genes of hlα,hlb,hlg and hld,and hemolysis test was conducted.Results Among the Staphylococcus aureus strains isolated from food,the detection rate of four kind of hemolysin genes(hlα,hlb,hlg and hld)were 55.9%,72.1%,75.7%and 83.8%respectively.The most common gene pattern were hlb/hlg/hld(33.1%).Among the Staphylococcus aureus strains isolated from healthy carriers,the detection rate of 4 kinds of hemolysin genes(hlα,hlb,hlg and hld)were 78.8%,51.3%,87.5%and 85.0%respectively.The most common gene pattern was hlα/hlg/hld(37.5%).The detection rate of hlb from dairy products was higher than that from rice wheat products(P<0.01).The detection rate of hlαfrom catering employees was higher than that from production workers of food company(P<0.05).The detection rate of complete hematolysis which isolated from employees was significantly higher than that isolated from food(P<0.05).Conclusion The detection rate and distribution pattern of hemolysin genes of Staphylococcus aureus isolated from different samples are different in statistic.The construction of hemolysin gene monitoring system should be strengthened.
