摘要
目的 运用网络药理学方法探讨红景天治疗脑出血的潜在作用机制。方法 通过TCMID数据库与文献检索收集红景天的化学成分,利用Swiss ADME数据库与Swiss Target Prediction数据库进行有效化合物筛选与靶点预测,通过Gene Cards和OMIM数据库获得脑出血相关靶点,交集法获取红景天—脑出血共同靶点,通过STRING数据库构建蛋白—蛋白相互作用(PPI)网络,利用Cytoscape软件构建红景天成分—靶点互作网络,进行可视化处理,并筛选节点度值较高的关键成分和核心靶点,通过Metascape数据库进行GO功能和KEGG通路富集分析,预测作用机制。结果 筛选出红景天治疗脑出血的有效活性成分32个,作用靶点142个,经分析获得AKT1、VEGFA、EGFR、MAPK3、STAT3、HIF1α、MMP-9、PTGS2等15个核心靶标可能是参与脂质与动脉粥样硬化、神经活性配体—受体相互作用、cGMP-PKG信号通路、p53信号通路及NF-κB信号通路发挥抑制细胞凋亡、调控炎性反应及调节血管生成等作用。结论 红景天通过多种不同成分作用于多个不同靶点,且协同调控多种不同信号通路,从整体网络层面发挥治疗脑出血作用。
Objective To investigate the potential mechanism of rhodiola rosea in the treatment of cerebral hemorrhage by using the method of network pharmacology.Methods The chemical constituents of rhodiola rosea were collected by TCMID database and literature retrieval, Swiss ADME database and Swiss Target Prediction database were used to screen effective compounds and predict targets, the targets related to cerebral hemorrhage were obtained by Gene Cards and OMIM database, the common targets of rhodiola rosea and intracerebral hemorrhage were obtained by intersection method,protein-protein interaction(PPI) network was constructed by STRING database, and the composition-target interaction network was constructed by Cytoscape software for visual processing, and the key components and core targets with high node degree values were screened, GO function and KEGG pathway enrichment were analyzed by Metascape database to predict the mechanism of action.Results A total of 32 active components and 142 targets of rhodiola rosea in the treatment of cerebral hemorrhage were screened out. Fifteen core targets, including AKT1, VEGFA, EGFR, MAPK3, STAT3, HIF1α, MMP-9and PTGS2, were found to be involved in the inhibition of lipid-atherosclerosis, neuroactive ligand receptor interaction,CPMP-PKG signaling pathway, p53 signaling pathway and NF-κB signaling pathway apoptosis, regulation of inflammatory response and regulation of angiogenesis.Conclusion Rhodiola rosea can act on multiple different targets through a variety of different components, and coordinately regulate variety of different signaling pathways, to play a role in the treatment of cerebral hemorrhage from the overall network level.
