响应面法优化一叶兰总皂苷超声提取工艺

Optimization of ultrasonic extraction process of total saponins from Aspidistra elatior Blume.by response surface methodology

摘要 以一叶兰根状茎为原料,以一叶兰总皂苷得率为指标,在单因素的基础上通过响应面法优化一叶兰总皂苷的提取工艺,并通过测定不同年份一叶兰生物量和总皂苷含量,构建一叶兰生物量增长模型及分析生物量与总皂苷含量的关系。结果表明:一叶兰总皂苷最佳提取工艺条件为乙醇体积分数68.32%、液料比27.58∶1(mL/g)、提取温度68.6℃、提取时间20 min,在此条件下总皂苷得率为17.0%;一叶兰总皂苷含量与生物量呈显著正相关(P<0.05);对不同年份一叶兰根状茎生物量进行Logistic模型拟合(R~2=0.778),模型预测1~5年一叶兰根状茎平均生物量分别为5.57、7.74、9.93、11.80、13.20 g。 Using the rhizome of Aspidistra elatior Blume.as raw material and the yield of total saponins of Aspidistra elatior Blume.as index,the extraction process was optimized by response surface method on the basis of single factor.By measuring the biomass and total saponins content of Aspidistra elatior Blume.in different years,the biomass growth model of Aspidistra elatior Blume.was constructed and the relationship between biomass and total saponins content was analyzed.The results showed that the optimum extraction conditions of total saponins from Aspidistra elatior Blume.were ethanol volume fraction 68.32 %,liquid solid ratio 27.58 ∶1(mL/g),extraction temperature 68.6 °C and extraction time 20 mins.Under these conditions,the yield of total saponins was 17.0 %.The content of total saponins was significantly positive correlated with biomass(P<0.05).A logistic model(R~2=0.778) was fitted to the rhizome biomass of Aspidistra elatior in different years,and the model predicted that the average rhizome biomass of Aspidistra elatior from 1 to 5 years was 5.57,7.74,9.93,11.80 and 13.20 g,respectively.
