摘要
为研究棉花黄萎病大丽轮枝菌Verticillium dahlia V991孢内毒素与棉花叶盘互作过程中叶绿素的荧光值变化,了解抗/感棉花-毒素互作与叶绿素荧光参数变化的关系,建立快速筛选抗黄萎病棉花品种的方法。以不同pH、不同温度优化大丽轮枝菌V991孢内毒素的提取,初期将获得的毒素与5个不同棉花品种的棉叶盘进行浸泡侵染,采用叶绿素荧光成像系统监测棉花叶圆盘在离体条件被孢内毒素侵染后天变化的荧光参数及图像成像变化,并用分生孢子水培法进一步验证,从而建立棉花黄萎病抗性鉴定方法,随后对18个棉花品种进行叶盘毒素侵染与分生孢子水培法鉴定,进一步确定抗/感棉花-毒素互作时与叶绿素荧光参数变化的相关性。结果显示:具有最强致病力的V991孢内毒素适宜温度为10℃,pH为6.5。抗/感棉花-毒素互作与叶绿素荧光参数实际量子产量Y(Ⅱ)、非光化学淬灭NPQ、光系统Ⅱ非调节性能量耗散的量子产量Y(NO)天变化趋势存在相关性,其中抗病品种Y(Ⅱ)、NPQ、Y(NO)这3个荧光参数天变化趋势明显,在11 d的调查中,分别呈现紧凑的倒勺状、“W”和“M”型,而感病品种荧光参数Y(Ⅱ)呈现的天变化趋势线无明显的形状,并且NPQ和Y(NO)这两个荧光的天变化趋势线呈现松散的“W”型及“M”型,同时利用该方法获得的各棉花抗病性结果与分生孢子水培法得到的抗病性结果一致。利用上述方法从23个不同的棉花品种(系)获得1个抗病品系,2个耐病品种(系)。综上可见,大丽轮枝菌孢内毒素叶盘侵染联合叶绿素荧光参数Y(Ⅱ)、NPQ、Y(NO)的持续测定,并根据叶绿素各荧光参数所形成的天变化趋势,能快速准确鉴定到抗/耐黄萎病棉花品种。
In order to study the changes of chlorophyll fluorescence during the interaction between the endotoxin from V.dahliae V991 and cotton leaf discs,and to explore the relationship between cotton-toxin and chlorophyll fluorescence parameters,a rapid method for screening cotton varieties resistant to Verticillium wilt was established.The extraction of endotoxins from V.dahliae V991 conidia was optimized by different pH and temperatures,and the cotton detached leaf discs of five different cotton varieties were soaked and inoculated with the obtained toxins.The changes of imaging and parameters were measured by chlorophyll fluorescence imaging system after the cotton leaf discs were infected with endotoxins in vitro,which was further verified by conidia hydroponics.The method was finally established for identifying cotton resistance to Verticillium wilt.Then toxin identification and hydroponic inoculation were used to detect the disease resistance of 18 cotton varieties,so as to further clarify the correlation between the interaction of resistant/susceptible cotton-toxin and the change of chlorophyll fluorescence.The results showed that the optimum temperature for the extraction of endotoxin from V991 conidia was 10℃and pH was 6.5.A clear correlation had been shown between resistant/susceptible cotton-toxin and day-to-day changes in fluorescence parameters,which included the maximum quantum yield F_(v)/F_(m),the actual quantum yield Y(Ⅱ),the non-photochemical quenching NPQ,and the quantum yield Y(NO),especially,in the disease-resistance varieties,the trends separately exhibited compact inverted spoon shape,‘W’and‘M’trend in the 11-day investigation.However,the day-to-day variation trend line of fluorescence parameter Y(Ⅱ)of susceptible varieties showed no obvious shape,and that of NPQ and Y(NO)presented loose‘W’and‘M’types.At the same time,the results of the disease resistance obtained with this method were consistent with the results of the disease index identification obtained with the conidia ro
