摘要
为了研究白术多糖对南海麻黄鸡免疫马立克氏病疫苗后的影响,试验将160只健康、体重相近的1日龄南海麻黄鸡随机分成4组,分别为对照组、马立克氏病疫苗组、白术多糖组和马立克氏病疫苗白术多糖组,每组4个重复,每个重复10只。预试1周后对照组和马立克氏病疫苗组只饲喂基础日粮,白术多糖组和马立克氏病疫苗白术多糖组在基础日粮中按体重添加200 mg/kg的白术多糖;马立克氏病疫苗组与马立克氏病疫苗白术多糖组均在8日龄和15日龄肌肉注射鸡马立克氏病活疫苗,对照组与白术多糖组未进行免疫。于试验开始前和开始后的第7,14,21,28天试验鸡以重复为单位称重,计算日增重,同时于试验开始后的第7,14,21,28天采集脾脏、胸腺、法氏囊、新鲜血液,分析脾脏指数、胸腺指数和法氏囊指数,制作切片观察第14,21,28天时的免疫器官组织形态变化,检测细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)、白细胞介素-1β(IL-1β)、γ干扰素(IFN-γ)水平。结果表明:试验第28天时,白术多糖组脾脏指数显著高于对照组(P<0.05),马立克氏病疫苗白术多糖组的法氏囊指数也高于马立克氏病疫苗组;白术多糖组与马立克氏病疫苗白术多糖组脾小结直径增大,且试验第21天时马立克氏病疫苗白术多糖组脾小结直径显著大于马立克氏病疫苗组(P<0.05)。第28天时白术多糖组淋巴滤泡面积显著大于对照组(P<0.05);与马立克氏病疫苗组比较,马立克氏病疫苗白术多糖组鸡血清中TNF-α、IL-2、IFN-γ含量有不同程度的升高,IL-1β含量降低;白术多糖组和马立克氏病疫苗白术多糖组全期日增重均大于对照组和马立克氏病疫苗组,且白术多糖组与对照组差异极显著(P<0.01)。说明白术多糖能够促进南海麻黄鸡免疫器官组织发育,增强免疫细胞分泌IL-2、TNF-α、IFN-γ的能力,对免疫马立克氏病疫苗后的雏鸡在免疫�
In order to study the effect of Atractylodes macrocephala polysaccharides on immunity of Marek’s disease(MD) vaccine in Nanhai Yellow chickens, in the experiment, 160 healthy 1-day-old Nanhai Yellow chickens with similar body weight were randomly divided into four groups, including control group, MD vaccine group, Atractylodes macrocephala polysaccharides group and MD vaccine with Atractylodes macrocephala polysaccharides group, with four replicates in each group, and 10 chickens in each replicate. After 1 week of pre-feeding, the control group and the MD vaccine group were only fed the basal diet, and the Atractylodes macrocephala polysaccharides group and the MD vaccine with Atractylodes macrocephala polysaccharides group were supplemented with 200 mg/kg of Atractylodes macrocephala polysaccharides according to body weight in the basal diet. MD vaccine group and MD vaccine with Atractylodes macrocephala polysaccharides group were injected intramuscularly with chicken MD live vaccine at the age of 8 and 15 days, while the control group and Atractylodes macrocephala polysaccharides group were not immunized. Daily gain was calculated by weighing chickens in replicate before and on the 7 th, 14 th, 21 st, and 28 th days after the start of the experiment;the spleen, thymus, bursa, and fresh blood were collected on the 7 th, 14 th, 21 st, and 28 th days of the experiment;analysis of spleen index, thymus index and bursa index was carried out;the sections were made to observe the morphological changes of the above-mentioned immune organs on the 14 th, 21 st, and 28 th days;the levels of cytokines tumour necrosis factor-α(TNF-α),interleukin-2(IL-2),interleukin-1β(IL-1β),and interferon-γ(IFN-γ) were detected. The results showed that on the 28 th day of the experiment, the spleen index of the Atractylodes macrocephala polysaccharides group was significantly higher than that of the control group(P<0.05);the bursa index of the MD vaccine with Atractylodes macrocephala polysaccharides group was also higher than that
