lncRNALINC01410靶向miR-205-5p抑制胰腺癌细胞放疗敏感性的研究 被引量:1

Research of lncRNA LINC01410 targeting miR-205-5p to inhibit the radiosensitivity of pancreatic cancer cells

摘要 目的探究长链非编码RNA(long noncoding RNA,lncRNA)LINC01410对miR-205-5p的靶向关系,及其对胰腺癌细胞放疗敏感性的影响。方法将SW1990-R细胞分为NC组(正常培养)、si-con组(转染si-con)、si-LINC01410组(转染si-LINC01410)、miR-con组(转染miR-con)、miR-205-5p组(转染miR-205-5p模拟物)、si-LINC01410+anti-miR-con组(共转染si-LINC01410和anti-miR-con)、si-LINC01410+anti-miR-205-5p组(共转染si-LINC01410和anti-miR-205-5p)。用荧光定量聚合酶链反应检测LINC01410与miR-205-5p的表达水平。4 Gy X线照射后,将细胞分为NC+4 Gy组、si-con+4 Gy组、si-LINC01410+4 Gy组、miR-con+4 Gy组、miR-205-5p+4 Gy组、si-LINC01410+anti-miR-con+4 Gy组、si-LINC01410+anti-miR-205-5p+4 Gy组。用流式细胞仪测定细胞的凋亡情况,用双荧光素酶报告基因法分析LINC01410和miR-205-5p的靶向关系。结果培养48 h后,si-con组和si-LINC01410组的LINC01410表达水平分别为0.97±0.12和0.37±0.06,miR-205-5p表达水平分别为1.05±0.16和3.09±0.36,差异均有统计学意义(均P<0.01)。miR-con组和miR-205-5p组的miR-205-5p表达水平分别为1.03±0.17和3.02±0.35,si-LINC01410+anti-miR-con组和si-LINC01410+anti-miR-205-5p组的miR-205-5p表达水平分别为3.06±0.35和1.39±0.18,差异均有统计学意义(均P<0.01)。si-con+4 Gy组和si-LINC01410+4 Gy组的细胞凋亡率分别为(8.30±0.89)%和(25.20±3.15)%,miR-con+4 Gy组和miR-205-5p+4 Gy组的细胞凋亡率分别为(8.70±0.89)%和(26.10±3.09)%,si-LINC01410+anti-miR-con+4 Gy组和si-LINC01410+anti-miR-205-5p+4 Gy组的细胞凋亡率分别为(24.60±2.56)%和(11.50±1.29)%,差异均有统计学意义(均P<0.01)。miR-205-5p是LINC01410的靶基因。结论LINC01410通过靶向miR-205-5p抑制胰腺癌细胞放疗敏感性。 Objective To explore the targeting relationship of long noncoding RNA(lncRNA)LINC01410 to miR-205-5 p and its effect on radiosensitivity of pancreatic cancer cells.Methods SW1990-R cells were divided into NC group(normal culture),si-con group(transfected with si-con),si-LINC01410 group(transfected with si-LINC01410),miR-con group(transfected with miR-con),miR-205-5 p group(transfected with miR-205-5p mimic),si-LINC01410+anti-mi R-con group(co-transfected with si-LINC01410 and anti-mi R-con),si-LINC01410+anti-mi R-205-5p group(co-transfected with si-LINC01410 and anti-mi R-205-5p).Fluorescence quantitative polymerase chain reaction was used to detect the expression levels of LINC01410 and mi R-205-5p.After 4 Gy X-ray irradiation,the cells were divided into NC+4 Gy group,si-con+4 Gy group,si-LINC01410+4 Gy group,mi R-con+4 Gy group,mi R-205-5p+4 Gy group,si-LINC01410+anti-mi R-con+4 Gy group,si-LINC01410+anti-mi R-205-5p+4 Gy group.Flow cytometry was used to measure cell apoptosis.The dual luciferase reporter gene method was used to analyze the targeting relationship between LINC01410 and mi R-205-5p.Results After treatment 48 h,the expression levels of LINC01410 in the si-con group and si-LINC01410 group were 0.97±0.12 and 0.37±0.06;the expression levels of mi R-205-5p were 1.05±0.16 and 3.09±0.36,respectively.All differences were statistically significant(all P<0.01).The expression levels of mi R-205-5p in mi R-con group and mi R-205-5p group were 1.03±0.17 and 3.02±0.35;the expression levels of mi R-205-5p in the si-LINC01410+anti-mi R-con group and si-LINC01410+anti-mi R-205-5p group were 3.06±0.35and 1.39±0.18,respectively,and the differences were statistically significant(all P<0.01).The apoptotic rates of si-con+4 Gy and si-LINC01410+4 Gy groups were(8.30±0.89)%and(25.20±3.15)%;the apoptotic rates of mi R-con+4 Gy and mi R-205-5p+4 Gy groups were(8.70±0.89)%and(26.10±3.09)%;the apoptotic rates of si-LINC01410+anti-mi R-con+4 Gy and si-LINC01410+anti-mi R-205-5p+4 Gy groups were(24.60±2.56)%and(1
