浓香白酒大曲中产普切明酸芽孢杆菌的筛选及其发酵条件优化 被引量:2

Screening and Fermentation Conditions Optimization of Pulcherrimin Acidic Acid-producing Bacillus licheniformis from Daqu of Chinese Strong Flavor Baijiu

摘要 普切明酸是一种由微生物产生的环二肽,因其螯合铁离子,具有较好的抑菌活性,可作为一种有效的生防制剂,在农业、医药和染料等领域具有潜在应用前景。然而,以微生物发酵方式产普切明酸,其产量水平仍然相对较低。从川南知名酒厂的大曲中筛选产普切明酸的芽孢杆菌菌株,获得1株地衣芽孢杆菌,对其发酵条件进行优化,根据单因素试验和正交试验得出最优发酵条件。结果表明,当温度40℃、接种量2%、pH 7.5、葡萄糖添加量40 g/L、磷酸氢二钾添加量0.5 g/L、(NH_(4))SO_(4)添加量8.6 g/L时,普切明酸产量达1 236 mg/L,较原始产量提高4.95倍。 Pulcherrimin acid is a cyclodipeptide produced by microorganisms. Because of its chelating iron ion and good antibacterial activity, Pulcherrimin acid can be used as an effective biocontrol agent and has potential application prospects in the fields of agriculture, medicine and dyes. However, the production level of Pulcherrimin acid by microbial fermentation is still low compared with other methods. Bacillus strains producing Pulcherrimin acid were screened from Daqu of wellknown distillery in southern Sichuan. The strain was Bacillus licheniformis, and its fermentation conditions were subsequently optimized. The optimal fermentation condition was obtained according to the single factor test and orthogonal test. The results showed that when the temperature was 40 ℃, inoculation amount was 2%, pH was 7.5, glucose was used as carbon source,and nitrogen source(NH_(4))SO_(4) was added at 8.6 g/L, the yield of Pulcherrimin acid reached 1 236 mg/L, which was 4.95 times higher than the original Pulcherrimin acid production.
