长期发酵大头菜差异代谢物及其关联代谢通路分析 被引量:2

Analysis of Differential Metabolites and Related Metabolism Pathways in Long-Term Fermented Kohlrabi Fermented for Different Periods

摘要 为探究工业化生产不同超长周期发酵大头菜代谢物的差异及其主要关联代谢通路,采用液相色谱-串联质谱技术结合多元统计分析,测定并比较0、5、10 a大头菜的代谢物。结果表明,发酵0 a与5 a、发酵5 a与10 a、发酵0 a与10 a分别鉴定出53、113种和174种差异代谢物(变量投影重要性值>1.3,P<0.05),主要为有机酸、氨基酸及核酸及其衍生物等。3个处理组共同的差异代谢物有19种,包含L-苯丙氨酸、β-D-果糖-6-磷酸等。差异代谢物京都基因与基因组百科全书通路富集分析显示,苯丙氨酸代谢、磷酸戊糖途径、果糖和甘露糖代谢途径与主要差异代谢物相关性最显著。本研究揭示了不同超长周期发酵大头菜代谢物的差异和主要差异代谢物及其关联代谢通路,为定向调控大头菜特征代谢物的生物合成提供一定理论依据。 In order to explore the differences in the metabolite profiles of industrially long-term fermented kohlrabi with different fermentation periods and the major metabolic pathways involved,the metabolite profiles of kohlrabi fermented for 0,5 and 10 years were analyzed and compared by using liquid chromatography-tandem mass spectrometry(LC-MS/MS)combined with multivariate statistical analysis.The results showed that a total of 53,113 and 174 differential metabolites were identified in years 0 versus 5,5 versus 10,and 0 versus 10 of fermentation,respectively(variable importance in the projection(VIP)>1.3,P<0.05),including organic acids,amino acids,nucleic acids and their derivatives.Nineteen differential metabolites were common to the three fermentation time points,including L-phenylalanine,andβ-D-fructose-6-phosphate.The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis of the differential metabolites showed that phenylalanine metabolism,the pentose phosphate pathway,and the fructose and mannose metabolic pathways were the most significantly correlated with the major differential metabolites.The present study provides a theoretical basis for targeted regulation of the biosynthesis of the characteristic metabolites in fermented kohlrabi.
