摘要
目的 探究TC2N表达在胶质瘤中的临床病理意义及其下游可能的信号通路,并分析TC2N成为抗癌治疗的潜在治疗靶点的可能性。方法基于两个公共数据库,Oncomine和The Cancer Genome Atlas(TCGA)对TC2N在胶质瘤中的表达进行了分析;单因素和多因素回归分析TC2N表达与胶质瘤患者预后的关系。利用基因集合富集分析(GSEA)和共表达网络用于探究TC2N下游可能的信号通路。结果 发现TC2N在肿瘤组织中表达显著高于非肿瘤组织。对TC2N表达与临床病理参数进行分析发现,TC2N表达与放疗史(n=530,P=0.010)和组织学类型(n=530,P<0.001)显著相关。TC2N高表达与胶质瘤患者预后呈负相关,尤其是对于有放疗史的胶质瘤患者,但与无放疗史的胶质瘤患者预后没有相关性,TC2N表达与TLR4信号通路呈正相关。结论 TC2N在胶质瘤中高表达,是胶质瘤患者生存预后的一个独立预后标志物。
Objective To explore the clinicopathological significance of TC2 N expression in glioma and its downstream possible signaling pathways, and analyze the possibility of TC2 N becoming a potential therapeutic target for cancer treatment.Methods The expression of TC2 N in glioma was analyzed based on two public databases: Oncomine and The Cancer Genome Atlas(TCGA).The relationship between TC2 N expression and prognosis of patients with glioma was analyzed by univariate analysis and multivariate regression analysis.Gene set enrichment analysis(GSEA) and co-expression network were used to explore the possible signaling pathways of TC2 N downstream.Results It was found that the expression of TC2 N in tumor tissues was significantly higher than that in non-tumor tissues.Analysis of TC2 N expression and clinicopathological parameters revealed a significant correlation of TC2 N expression with radiotherapy(n=530,P=0.010) and histological type(n=530,P<0.001).The high expression of TC2 N was negatively correlated with the prognosis of glioma patients, especially for glioma patients with radiotherapy history, but was not correlated with the prognosis of glioma patients without radiotherapy history.The expression of TC2 N was positively correlated with TLR4 signaling pathway.Conclusion TC2 N is highly expressed in glioma and is an independent prognostic marker for the survival and prognosis of patients with glioma.
