摘要
以C.glutamicum中的NCgl0877(命名为CgAhp)为研究对象,揭示了CgAhp内在生理表型及其发挥这些功能的生化特性和调控机制.结果表明:C.glutamicum中cgahp基因缺失突变体仅对有机过氧化物异丙苯过氧化氢(CHP)和叔丁基过氧化氢(t-BHP)高度敏感,对无机氧化剂、烷化剂、抗生素、重金属不敏感,说明CgAhp在维持C.glutamicum抗有机氧化应激方面发挥重要作用;CgAhp主要协同二氢脂肪酰胺脱氢酶(Lpd)/二氢脂肪酰胺琥珀酰转移酶(SucB)/NADH电子转移途径发挥氧化还原酶的作用.研究发现CgAhp的特性与AhpD类似,是有机氧化应激耐受性的关键二硫氧化还原酶和过氧化物酶的功能性电子供体.
This paper considers the physiological and biochemical functions of NCgl0877 which is designated CgAhp in C.glutamicum.The cgahp-deleted strain is highly sensitive to the organic peroxides cumene hydroperoxide(CHP) and tertbutyl hydroperoxide(t-BHP),but not sensitive to inorganic oxidants,including alkylating agents,antibiotics,and heavy metals,indicating that CgAhp plays an important role in maintaining the resistance of C.glutamicum to organic oxidative stress.CgAhp acts as an oxidoreductase preferentially linked to the dihydrolipoamide dehydrogenase(Lpd)/dihydrolipoamide succinyltransferase(SucB)/NADH system.In summary,this study identifies CgAhp,behaving like AhpD,as a key disulfide oxidoreductase involved in the oxidative stress tolerance and the functional electron donor for peroxidase.
