摘要
为了证实刺山柑凝胶对类风湿性关节炎大鼠模型的治疗作用及机理,本研究建立了弗氏完全佐剂关节炎大鼠模型,经关节炎评分标准评价将造模成功大鼠随机分为刺山柑凝胶(高、中、低剂量)组、模型组、阳性药组,并给予1次/d,连续30d的药物干预。过程中记录大鼠体征状态、体重及足趾肿胀度;疗程结束后测量免疫器官指数,制作切片,分别HE(苏木精-伊红染色法)、甲苯胺蓝染色观察大鼠踝关节病理组织形态学变化;测定血清中细胞因子TNF-α、IL-1β、IL-17、IL-10表达水平。经各项观察指标组间比较结果证实,与模型组比较,刺山柑凝胶高、中、低剂量组明显抑制足趾肿胀度,促进免疫器官发育、增强机体免疫,发挥免疫调节作用;减少关节组织的炎细胞浸润、改善组织病理形态;下调TNF-α、IL-1β、IL-17,上调IL-10表达水平,差异均具有统计学意义(P<0.01)。综上,刺山柑凝胶经局部病灶部位皮肤给药后,能通过下调促炎因子TNF-α、IL-1β、IL-17和上调抑炎因子IL-10表达水平作用机理,达到缓解类风湿性关节炎症状、缩短病程及治疗疾病的作用。
In order to prove the therapeutic effect and mechanism of Capparis spinosa L.gels on rheumatoid arthritis rats,a Freund’s adjuvant arthritis rat model was established.The rats were randomly divided into three groups:the Capparis Spinosa L.Gels treatment groups(high,medium and low doses groups),the model group and the positive drug group were given 1 time/d and continuous 30d intervention.The physical signs,body weight and toe swelling were recorded.After the course of treatment,the immune organ index was measured,sections were made,and the pathomorphological changes of rat ankle were observed by HE and toluidine blue staining;The expression levels of TNF-α、IL-1 βIL-17 and IL-10 in serum was measured.The results showed that the high,medium and low dose groups of Capparis spinosa L.gels significantly inhibited the swelling of the toes,promoted the development of immune organs,enhanced the immunity of the body,and played an immunoregulatory role,reduced inflammatory cell infiltration of joint tissue and improved histopathological morphology,comparing with the model group.Down-regulated the expression level of TNF-α、IL-1 β、 IL-17,up-regulated the expression level of IL-10,the difference was statistically significant(P<0.01).In conclusion,after local lesion skin administration,by down-regulating the expression level of proinflammatory factor TNF-α、IL-1β and IL-17,and up-regulating the expression level of anti-inflammatory factor IL-10,Capparis spinosa L.gels could alleviate the symptoms of rheumatoid arthritis,as well as shorten the course of disease and treat the disease.
