N、P、K平衡施肥对金银花产量和品质及相关基因表达的影响 被引量:5

Effects of the balanced fertilization of N,P and K on the yield,quality and related gene expression in Lonicera japonica

摘要 【目的】通过N、P、K平衡施肥提高金银花产量和品质,分析其促进绿原酸和木犀草苷积累的机制。【方法】以金银花“北花一号”3 a生植株为材料,施用4个不同比例N、P、K[CK(1∶1∶1)(对照)、T(11∶2∶1)、T(22∶1∶1)和T(31∶1∶2)],测定二白期花蕾产量和品质及相关基因表达水平。【结果】N、P、K平衡施肥可显著促进植株的生长发育(如着花节数、花朵鲜重和花蕾数),提高花蕾产量、主要活性物质(绿原酸、木犀草甘、可溶性糖、总黄酮类和总酚类化合物)含量、抗氧化能力,诱导绿原酸和木犀草苷生物合成途径中7个关键酶基因(PAL3、C4H2、4CL1、HQT1、CHS2、CHI2和FNS1)的表达;其中,以T2平衡施肥效果最佳,相比CK、T1和T3,产量分别提高1.98、1.29和1.21倍,绿原酸、木犀草苷、可溶性糖、总黄酮、酚类化合物含量和抗氧化能力最大提高1.79、1.71、1.80、1.64、1.92和1.24倍,7个关键基因PAL3、C4H2、4CL1、HQT1、CHS2、CHI2和FNS1的表达水平最大上调7.00、7.10、3.96、4.58、2.07、1.51和3.27倍。【结论】N、P、K为2∶1∶1的平衡施肥可显著提高金银花的产量和品质,通过上调相关基因的表达可促进阿魏酸和木犀草苷的合成和积累。 【Objective】The study aimed to improve the yield and quality of Lonicera japonica by the bal‐anced fertilization of N,P and K,and to explore the mechanism of chlorogenic acid and galuteolin accumulation in L.japonica.【Method】The 3-year-old plants of L.japonica Beihua No.1 were employed and four fertilization treatments of different ratio of N,P and K were performed,i.e.,CK(1∶1∶1),T1(1∶2∶1),T2(2∶1∶1)and T3(1∶1∶2),followed by measuring the yield and quality of flower buds and by analyzing related genes expression at the two-white flowering stage.【Result】The balanced fertilization of N,P and K significantly enhanced plant growth and development(e.g.,the number of knots with buds,fresh weight of flower buds,and flower bud number),improved flower bud yield and the content of main bioactive compounds(e.g.,chlorogenic acid,cynaroside,soluble sugar,total flavonoids,and total phenols),and the antioxidant capacity.In addition,the balanced fertilization could induce the up-regulation expression of seven genes(i.e.,PAL3,C4H2,4CL1,HQT1,CHS2,CHI2 and FNS1)involved in biosynthesis of chlorogenic acid and galu‐teolin.Among the four treatments,the T2 showed the best effectiveness in increasing the yield,which in‐creased by 1.98,1.29 and 1.21 fold,respectively,compared to CK,T1 and T3;the levels of chlorogenic acid,galuteolin,soluble sugar,total flavonoids,total phenols and antioxidant capacity increased by 1.80,1.64,1.92,1.79,1.71and 1.24 fold,respectively;and the expression levels of PAL3,C4H2,4CL1,HQT1,CHS2,CHI2 and FNS1 were regulated up to 7.00,7.10,3.96,4.58,2.07,1.51 and 3.27 fold,re‐spectively.【Conclusion】The balanced fertilization of N,P and K with the ratio of 2∶1∶1 could significantly improve the yield and quality of L.japonica,and could enhance the accumulation of chlorogenic acid and galuteolin by up-regulating related gene expression.These findings could be used as scientific reference to the rational fertilization in production of L.japonica.
