糖尿病大鼠睾丸损伤疾病模型的建立和评价 被引量:1

Establishment and evaluation of testicular injury disease model in diabetic rats

摘要 目的研究实验性糖尿病SD大鼠睾丸损伤模型建立的方法,并对其有效性进行评估。方法将8周龄SD大鼠随机分为正常组和模型组,每组10只。正常组给予等量柠檬酸缓冲液处理,模型组给予一次性空腹腹腔注射45 mg·kg^(-1)链脲佐菌素。7周后,比较随机血糖、睾丸指数、抗氧化因子水平、睾丸组织损伤的变化。结果模型组和正常组的随机血糖分别为(25.60±3.34)和(5.30±0.62)mmol·L^(-1),超氧化物歧化酶分别为(25.56±3.38)和(69.00±5.86)U·mL^(-1),谷胱甘肽分别为(8.56±6.10)和(11.56±3.92)mmol·L^(-1),过氧化氢酶分别为(35.89±3.78)和(43.20±2.13)U·mL^(-1),丙二醛分别为(13.93±3.81)和(11.34±1.30)nmol·mL^(-1)。模型组的上述指标与正常组比较,差异均有统计学意义(均P<0.05)。结论空腹腹腔一次性快速注射45 mg·kg^(-1)链脲佐菌素能引起大鼠睾丸功能紊乱及糖尿病睾丸损伤典型病理改变,成功建立糖尿病大鼠睾丸损伤模型。 Objective To study the method of establishing testicular injury model in experimental diabetic SD rats and evaluate its effectiveness.Methods Eight-week-old SD rats were randomly divided into normal group and model group with 10 rats in each group.The normal group was treated with citric acid buffer solution,and the model group was given a one-time fasting intraperitoneal injection of 45 mg·kg^(-1) streptozotocin.After 7 weeks,the changes of random blood glucose,testicular index,antioxidant factor level and testicular tissue damage were compared.Results The random blood glucose of model group and normal group were(25.60±3.34)and(5.30±0.62)mmol·L^(-1),super oxide dimutese were(25.56±3.38)and(69.00±5.86)U·mL^(-1),L-glutathione were(8.56±6.10)and(11.56±3.92)mmol·L^(-1),catalase were(35.89±3.78)and(43.20±2.13)U·mL^(-1),malondialdehyde were(13.93±3.81)and(11.34±1.30)nmol·mL^(-1),respectively.There were statistically significant differences between model group and normal group(all P<0.05).Conclusion Rapid one-time injection of 45 mg·kg^(-1) streptozotocin into the abdominal cavity can induce testicular dysfunction and typical pathological changes of diabetic testicular injury in rats,and a model of testicular injury in diabetic rats was established successfully.
