隔药饼灸对动脉粥样硬化易损斑块兔Beclin1、LC3蛋白表达的影响 被引量:1

Effects of Herbal-cake-separated Moxibustion on the Expressions of Beclin1 and LC3 Protein in Rabbits with Atherosclerotic Vulnerable Plaques

摘要 目的观察隔药饼灸对动脉粥样硬化(AS)易损斑块兔Beclin1、LC3蛋白表达的影响,探究隔药饼灸稳定AS易损斑块的作用机制。方法将58只雄性新西兰大耳白兔随机分为空白组、模型组、隔药饼灸组、直接灸组和西药组。采用高脂饲料喂养+腹主动脉球囊损伤+基因转染+药物触发斑块破裂制备AS易损斑块兔模型,分别予相应干预8周。采用ELISA检测血清总胆固醇(TC)、三酰甘油(TG)含量,透射电镜观察腹主动脉超微结构及自噬体形成情况,Westernblot检测腹主动脉Beclin1、LC3蛋白表达。结果与空白组比较,模型组兔血清TC、TG含量显著增加,腹主动脉组织Beclin1蛋白表达显著升高(P<0.01);与模型组比较,隔药饼灸组和西药组兔血清TC和TG含量显著减少(P<0.01,P<0.05),腹主动脉组织Beclin1、LC3Ⅱ蛋白表达及LC3Ⅱ/LC3Ⅰ比值显著升高(P<0.01,P<0.05);与直接灸组比较,西药组兔腹主动脉组织Beclin1蛋白表达显著降低(P<0.01)。透射电镜结果显示,模型组兔腹主动脉泡沫细胞增多,核膜、细胞器严重损伤,未见自噬体形成;直接灸组、隔药饼灸组兔腹主动脉泡沫细胞增多,核膜边界不清,细胞器损坏程度未见明显改变,可见自噬体形成;西药组兔腹主动脉泡沫细细胞损伤程度减轻,部分细胞器及胶原纤维保留,未见自噬体形成。结论隔药饼灸可能通过上调Beclin1、LC3Ⅱ蛋白表达,提高细胞自噬水平,促进自噬体形成,稳定AS易损斑块。 Objective To observe the effects of herbal-cake-separated moxibustion on the expressions of Beclin1 and LC3 in abdominal aorta of rabbits with atherosclerotic vulnerable plaques;To explore its mechanism in stabilizing vulnerable plaques.Methods Totally 58 male New Zealand white rabbits were randomly divided into blank group,model group,herbal-cake-separated moxibustion group,direct moxibustion group and Western medicine group.Atherosclerotic vulnerable plaques model was established by high-fat feeding+abdominal aortic balloon injury+gene transfection+triggered plaques rupture.Each group was given corresponding interventions for 8 weeks.ELISA was used to detect the contents of total cholesterol(TC),triacyl glycerin(TG)in serum;transmission electron microscope was used to to observe the ultrastructure of abdominal aorta and autophagosomes;Western blot was used to to detect the expressions of Beclin1 and LC3 protein in abdominal aorta.Results Compared with the blank group,the contents of TC and TG in serum of rabbits in the model group significantly increased,the proteins expression of Beclin1 in abdominal aorta significantly increased(P<0.01).Compared with the model group,the contents of TC and TG in serum of the rabbits in herbal-cake-separated moxibustion group and Western medicine group decreased(P<0.01,P<0.05),the proteins expression of Beclin1,LC3Ⅱand the ratio of LC3Ⅱ/LC3Ⅰsignificantly increased(P<0.01).Compared with the direct moxibustion group,the expression of Beclin1 protein in abdominal aorta of the Western medicine group significantly decreased(P<0.01).Transmission electron microscopy results showed that the number of foam cells in abdominal aorta of the model group increased,the nuclear membrane and organelles were damaged,and no autophagosomes were formed;the number of foam cells in abdominal aorta of the direct moxibustion group and herbal-cake-separated moxibustion group increased,the nuclear membrane boundary was unclear,the degree of organ damage did not change significantly,and the formatio
