摘要
目的建立巴马小型猪单侧药物性聋模型,为内耳疾病提供新的动物模型。方法选择6只健康巴马小型猪为实验动物,左耳为注射侧,右耳为对照侧,连续7天以硫酸新霉素100mg/kg行左侧乳突骨膜下注射,对照侧不做处理。观察实验前后小型猪一般情况、血肌酐、血尿素氮水平、听性脑干反应(Auditory brainstem response,ABR)以及耳蜗毛细胞形态。结果给药后,实验动物全部存活,血肌酐及尿素氮水平较给药前无统计学差异。给药后,双耳在24kHz、32kHz处ABR阈值均较给药前显著升高(P<0.01,t检验),在16kHz处注射侧ABR阈值较给药前有显著差异(P<0.01,t检验);注射侧耳ABR阈移均高于或等于对侧耳,且在16kHz、24kHz、32kHz处存在统计学差异(P<0.01,t检验)。给药后,小型猪注射侧ABR阈值在16kHz、24kHz、32kHz处显著高于对照侧(P<0.01,t检验)。耳蜗基底膜铺片可见注射侧耳蜗外毛细胞的纤毛结构受损重于对侧。结论单侧颞骨乳突骨膜下注射硫酸新霉素可造成小型猪双耳高频听力损伤,注射侧听阈显著高于对侧,是理想的单侧药物性聋动物模型。
Objective To establish a model of unilateral drug induced hearing loss in Bama miniature pig for research on inner ear diseases.Method Six healthy Bama miniature pigs received daily subperiosteal injection of neomycin sulfate at 100 mg/kg over the left mastoid for 7 days with the right ear as the control.The animals’general condition,blood levels of creatinine and urea nitrogen,ABRs and morphology of cochlear outer hair cells were examined.Result All animals survived drug administration and showed no significant increase in blood levels of creatinine and urea nitrogen.Upon completion of drug administration,ABR thresholds at 24 and 32 kHz were significantly higher on both the injection and control sides than before drug administration(P<0.01,t-test).ABR threshold was higher at 16,24 and 32 kHz on the injected side than the contralateral side(P<0.01,t-test).Outer hair cell cilia showed more damage on the injected side than on the control side.Conclusion Unilateral subperiosteal neomycin injection over the mastoid can cause high frequency hearing loss in both ears in miniature pigs,with significantly more loss on the injected side.This is a promising animal model of unilateral drug induced hearing loss.
