摘要
目的:建立藏荆芥中荆芥内酯的含量测定方法。方法:采用高效液相色谱法,色谱柱选择:Supersil ODS2 250 mm×4.6 mm,5μm,流动相为乙腈-水(体积比为40∶60),流速为1.0 mL/min,检测波长为216 nm,柱温为30℃。结果:荆芥内酯的线性关系方程为Y=12795176.3884X+607348.6713,R^(2)=0.9994,在0.01~5.0 mg/mL范围内的峰面积与进样浓度线性关系良好(R^(2)=0.9994)。精密度、重复性及稳定性实验的相对标准偏差(RSD)分别为1.46%、1.19%、1.83%,平均加样回收率为97.66%,RSD为1.56%。藏荆芥中荆芥内酯的含量为0.86%,不同药用部位中荆芥内酯含量差异明显。结论:建立的高效液相色谱法方法简单、准确、快速,重复性好,专属性强,可应用于藏荆芥中荆芥内酯的含量测定。
Objective: To establish a method for determination of Nepeta angustifolia C.Y.Wu. Methods: A high-performance liquid chromatography detection method was established. The column was Supersil ODS2 250 mm×4.6 mm, 5 μm and the mobile phase was acetonitrile-water(volume ratio 40∶60) at a flow rate of 1.0 mL/min. The detection wavelength was 216 nm, and the column temperature was 30 ℃. Results: The linear equation of nepetalactone was Y=12795176.3884X+607348.6713, and the peak area in the range of 0.01~5.0 mg/mL had a good linear relationship with the injection concentration(R^(2)=0.9994).The relative standard deviations(RSD) of precision, repeatability and stability experiments were 1.46 %, 1.19 % and 1.83 %, respectively. The average recovery rate was 97.66 % and RSD was 1.56 %. The content of nepetalactone in Nepeta angustifolia C.Y.Wu is 0.86 %, while which was obviously discrepant in different medicinal parts. Conclusion: The established HPLC method was simple, accurate, rapid, reproducible and specific, which could be applied to the determination of Nepeta angustifolia C.Y.Wu.
