摘要
目的探讨微小RNA(miR)-29a-3p影响Serpinh1对胃癌细胞的增殖与侵袭,并分析其相关的分子机制。方法检测miR-29a-3p在胃癌BGC823细胞中的表达水平,并评估miR-29a-3p在胃癌细胞增殖、迁移和侵袭中的作用。通过Western blotting和荧光素酶分析证明,miR-29a-3p与Serpinh13’UTR直接结合。此外,采用MTT法、集落形成法和Transwell法在体外检测miR-29a-3p/Serpinh1轴对胃癌细胞增殖、迁移和侵袭的影响。结果转染后miR-29a-3p模拟物(mimic)组中miR-29a-3p的表达明显高于miR-29a-3p阴性对照和空白组,转染miR-29a-3p mimic后BGC823细胞增殖能力显著降低。荧光素酶分析显示,miR-29a-3p通过靶向Serpinh13’-非翻译区(3’UTR)抑制Serpinh1的表达。此外,miR-29a-3p的过表达通过靶向Serpinh1显著抑制胃癌细胞的增殖、侵袭和迁移。结论MiR-29a-3p可靶向作用于Serpinh1,从而调控胃癌细胞的增殖与侵袭。
Objective To investigate the effects of microRNA(miR)-29a-3p on the proliferation and invasion of gastric cancer cells and analyze its related molecular mechanism.Methods The expression level of miR-29a-3p in gastric cancer cells was detected,and the role of miR-29a-3p in the proliferation,migration,and invasion of gastric cancer cells was evaluated.Western blotting and luciferase analysis showed that miR-29a-3p was directly bound to Serpinh13’-untranslated region(3’UTR).In addition,the effects of the miR-29a-3p/Serpinh1 axis on the proliferation,migration,and invasion of gastric cancer cells were detected by MTT assay,colony formation assay,and Transwell assay in vitro.Results After transfection,the expression of miR-29a-3p in the miR-29a-3p mimic group was significantly higher than that in the miR-29a-3p negative control and blank group.After transfection,the proliferation of BGC823 cells decreased significantly.Luciferase analysis showed that miR-29a-3p inhibited the expression of Serpinh1 by targeting the 3’UTR of Serpinh1.In addition,overexpression of miR-29a-3p significantly inhibited the proliferation,invasion,and migration of gastric cancer cells by targeting Serpinh1.Conclusion MiR-29a-3p can target Serpinh1 and regulate the proliferation and invasion of gastric cancer cells.
