摘要
目的建立同时测定毛茛中原儿茶酸、咖啡酸含量的HPLC方法,为评价毛茛药材质量提供一定参考。方法采用WondaSil^(TM) C_(18)-WR色谱柱(250 mm×4.6 mm,5μm),以乙腈(A)-0.1%磷酸溶液(B)为流动相,梯度洗脱,检测波长为261 nm(原儿茶酸)、324 nm(咖啡酸),柱温40℃,流速0.8 mL·min^(-1)。结果原儿茶酸和咖啡酸分别在0.84~84.00μg·mL^(-1)(r^(2)=0.9999)、0.78~78.08μg·mL^(-1)(r^(2)=1.0000)范围内线性关系良好;精密度、重复性、稳定性良好;平均加样回收率分别为96.11%(RSD=3.27%,n=6),94.68%(RSD=2.69%,n=6)。结论建立的方法简便、准确,可为毛茛药材质量评价提供参考。
OBJECTIVE To establish an HPLC method for simultaneous determination of protocatechuic acid and caffeic acid in Ranunculus japonicus to provide a certain reference for evaluating the quality of Ranunculus japonicus.METHODS HPLC experiment was performed on WondaSil^(TM) C_(18)-WR column(250 mm×4.6 mm,5μm)with mobile phase consisted of acetonitrile-0.1%phosphoric acid solution(gradient elute).The detection wavelength was 261 nm(for protocatechuic acid)and 324 nm(for caffeic acid).The flow rate was 0.8 mL·min^(-1) and the column temperature was maintained at 40℃.RESULTS Protocatechuic acid and caffeic acid showed good linear relationship in the range of 0.84-84.00μg·mL^(-1)(r^(2)=0.9999)and 0.78-78.08μg·mL^(-1)(r^(2)=1.0000),respectively.The precision,repeatability,and stability were good.The average recoveries of protocatechuic acid and caffeic acid were 96.11%(RSD=3.27%,n=6)and 94.68%(RSD=2.69%,n=6),respectively.CONCLUSION The established method is simple and accurate,which can provide reference for the quality evaluation of Ranunculus japonicus.
