摘要
目的探讨姜黄素对肝细胞癌耐药细胞HepG2/ADM上皮-间质转化(EMT)的影响及分子机制。方法取人肝细胞癌耐药细胞HepG2/ADM,以预实验确定的姜黄素合适浓度20μmol/L处理细胞。采用噻唑蓝溴化四唑法、划痕实验、Transwell实验分别检测姜黄素对细胞增殖、迁移、侵袭能力的影响,Western blot法检测姜黄素对EMT标志物及NF-κB/Snail通路相关蛋白表达的影响。结果相较于HepG2/ADM组,HepG2/ADM+姜黄素组细胞增殖、迁移和侵袭能力均明显减弱(均P<0.05),E-cadherin蛋白表达上调(P<0.05),N-cadherin、Vimentin、NF-κB p65、Snail蛋白表达均明显下调(均P<0.05)。HepG2/ADM细胞过表达Snail后,由姜黄素诱导的E-cadherin表达上调、N-cadherin和Vimentin表达下调均被逆转(均P<0.05),细胞增殖、迁移和侵袭能力均明显增强(均P<0.05)。结论姜黄素能够抑制HepG2/ADM细胞增殖、迁移和侵袭能力,并通过NF-κB/Snail通路抑制细胞的EMT过程。
Objective To investigate the effect and molecular mechanism of curcumin on epithelial-mesenchymal transition(EMT)of drug-resistant hepatocellular carcinoma cells.Methods Drug-resistant human hepatocellular carcinoma HepG2/ADM cells were treated with curcumin(20μmol/L).Cell proliferation,migration and invasion were detected with MTT,wound healing and Transwell assay,respectively.The expression of EMT markers and NF-κB/Snail pathway-related proteins was detected by Western blot.Results The proliferation,migration and invasion abilities of HepG2/ADM cells were significantly reduced after curcumin treatment(all P<0.05),the expression of E-cadherin protein was up-regulated(P<0.05),N-cadherin,Vimentin,NF-κB p65 and Snail protein expressions were significantly down-regulated(all P<0.05).While the up-regulation of E-cadherin expression and the down-regulation of N-cadherin and Vimentin expression induced by curcumin were reversed in Snail-overexpressed HepG2/ADM cells(all P<0.05),and the cell proliferation,migration and invasion abilities were significantly enhanced(all P<0.05).Conclusion Curcumin can inhibit the proliferation,migration and invasion,as well as the EMT of HepG2/ADM cells through NF-κB/Snail pathway.
