Expression of forkhead box protein A1 in breast cancer and its biological significance

摘要 目的探讨叉头框蛋白A1(FoxA1)在乳腺癌组织中表达及其对乳腺癌细胞增殖、周期和凋亡的影响。方法收集我院2019年1月至2022年1月的65例乳腺癌组织和癌旁组织作为研究对象, 采用免疫组织化学分析FoxA1蛋白表达水平;人乳腺癌细胞系MCF-7分为对照组和FoxA1 KD组, 分别采用慢病毒感染建立稳定细胞系, 采用细胞计数试剂盒(CCK-8)和克隆形成实验测定细胞增殖能力;采用流式细胞术分析两组细胞凋亡水平和周期变化;采用荧光定量聚合酶链反应(PCR)分析FoxA1对凋亡和周期相关蛋白的影响。组间数据比较采用t检验。结果癌旁组织中FoxA1蛋白相对表达水平(1.46±0.75)明显低于神经胶质瘤组织WDR12蛋白相对表达水平(2.66±0.57), 差异有统计学意义(t=10.280, P<0.05)。对照组细胞吸光度(A)值(1.91±0.06)明显高于FoxA1 KD组细胞A值(1.44±0.09), 差异有统计学意义(t=10.650, P<0.05)。对照组细胞克隆形成率[(80.55±3.60)%]明显高于FoxA1 KD组细胞克隆形成率[(42.67±2.51)%], 差异有统计学意义(t=21.160, P<0.05)。对照组细胞凋亡比例[(3.77±1.00)%]明显低于FoxA1 KD组细胞凋亡比例[(32.24±5.93)%], 差异有统计学意义(t=11.600, P<0.05)。对照组细胞G0/G1期比例[(33.74±2.59)%]低于FoxA1 KD组细胞G0/G1期比例[(41.52±2.52)%]比较, 差异无统计学意义(t=5.721, P>0.05)。对照组细胞S期比例[(33.20±2.29)%]明显高于FoxA1 KD组细胞S期比例[(26.65±2.08)%], 差异有统计学意义(t=5.181, P<0.05)。对照组和FoxA1 KD组细胞G2/M期比例[(34.77±2.95)%、(34.18±1.68)%]比较, 差异无统计学意义(t=0.431, P>0.05)。对照组细胞半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3 mRNA表达水平(0.92±0.09)明显低于FoxA1 KD组细胞(1.61±0.18), 差异有统计学意义(t=8.633, P<0.05)。对照组细胞周期蛋白D1(Cyclin D1) mRNA表达水平(0.95±0.04)明显高于FoxA1 KD组细胞(0.50±0.11), 差异有统计学意义(t=9.082, P<0.05)。结论 FoxA1� Objective To investigate the expression of forkhead box protein A1(FoxA1)in breast cancer and its effects on proliferation,cell cycle and apoptosis of breast cancer cells.Methods Totally,65 cases of breast cancer tissues and adjacent tissues collected in our hospital from January 2019 to January 2022 were collected as research objects.The expression level of FoxA1 protein was analyzed by immunohistochemistry.Human breast cancer cell line MCF-7 was divided into control group and FoxA1 KD group.The stable cell lines were established by lentivirus infection,and the cell proliferation ability was determined by cell counting kit-8(CCK-8)and in vitro tumor transplantation experiment.Flow cytometry was used to analyze the level and cycle of apoptosis in the two groups.The effects of FoxA1 on apoptosis and cycle related proteins were analyzed by fluorescence quantitative polymerase chain reaction(PCR).T-test was used to compare the data between groups.Results The relative expression level of FoxA1 protein in paracancerous tissues(1.46±0.75)was significantly lower than that of wdr12 protein in glioma tissues(2.66±0.57,t=10.280,P<0.05).The cell light absorption(A)value of the control group(1.91±0.06)was significantly higher than that of the FoxA1 KD group(1.44±0.09,t=10.650,P<0.05).The cell clone formation rate of control group[(80.55±3.60)%]was significantly higher than that of FoxA1 KD group[(42.67±2.51)%,t=21.160,P<0.05].The percentage of apoptosis in control group[(3.77±1.00)%]was significantly lower than that in FoxA1 KD group[(32.24±5.93)%,t=11.600,P<0.05].The G0/G1 phase ratio of cells in the control group[(33.74±2.59)%]was lower than that in the FoxA1 KD group[(41.52±2.52)%,t=5.721,P>0.05].The proportion of S phase cells in control group[(33.20±2.29)%]was significantly higher than that in FoxA1 KD group[(26.65±2.08)%,t=5.181,P<0.05].There was no significant difference in G2/M phase ratio between control group and FoxA1 KD Group[(34.77±2.95)%,(34.18±1.68)%,t=0.431,P>0.05].The expression level of cystei
