忍冬叶不同溶剂提取物与抗氧化的谱效关系研究 被引量:1

摘要 目的通过研究忍冬叶色谱峰与抗氧化活性之间的相关性分析,揭示忍冬叶抗氧化活性的可能的物质基础。方法采用HPLC-MS/MS建立忍冬叶11种不同极性提取物的指纹图谱,通过与对照品、文献信息比对、二级碎片信息指认共有峰对应的化合物。以DPPH自由基清除率及总还原力为指标,评价11种提取物抗氧化作用,通过灰色关联度法和偏最小二乘法联合分析共有峰与抗氧化作用的谱效关系,确认各共有峰的贡献程度。结果忍冬叶11种不同溶剂提取物中有20个共有峰,指认出18个化合物。S9清除DPPH自由基作用及总还原力活性均最强,其IC_(50)值分别为(1.08±0.15)mg·mL^(-1)、(3.41±0.10)mg·mL^(-1);18个化合物与药效的灰色关联度分析关联度均>0.8,除峰8、6、10、11外,其余峰与药效的偏最小二乘法分析回归系数均大于0,成正相关,其贡献程度排名前九位的共有峰为:峰5>4>16>17>18>20>19>1>9。结论忍冬叶中3,4-二羟基肉桂酸(峰5)、绿原酸(峰4)、3,4-二咖啡酰奎宁酸(峰16)、3,5-二咖啡酰奎宁酸(峰17)、4,5-二咖啡酰奎宁酸(峰18)、木犀草素(峰20)、槲皮素(峰19)、5-咖啡酰奎宁酸(峰1)、4-咖啡酰奎宁酸(峰9)可能是抗氧化的主要活性成分。 Objective To reveal the possible material basis of the antioxidant activities of Lonicera japonica leaves by studying the correlation between its chromatographic peak and its antioxidant activities.Methods The fingerprints of the 11 different polar extracts of Loniceras japonica leaves were established by HPLC-MS/MS,and the compounds corresponding to the common peaks were identified by comparing with the reference substance,literature information and secondary fragment information.Taking the DPPH radical scavenging rate and total reducing power as indicators,the antioxidant effects of the 11 extracts were evaluated.The spectrum-effect relationship between the common peaks and the antioxidant effects was analyzed by the grey correlation degree method and the partial least squares method to confirm the contribution degree of each common peak.Results There were 20 common peaks in the 11 different solvent extracts of Lonicola japonica leaves,and 18 compounds were identified.S9 had the strongest DPPH radical scavenging effect and total reducing power activity,and its IC_(50) values were(1.08±0.15)mg·mL^(-1) and(3.41±0.10)mg·mL^(-1) respectively.The grey correlation analysis of the 18 compounds and their efficacy showed that the correlation degree was greater than 0.8.Except the peaks 8,6,10 and 11,the remaining peaks were positively correlated with the regression coefficients of the partial least squares analysis of their efficacy,and the nine common peaks with the top contribution were 5>4>16>17>18>20>19>1>9.Conclusion The 3,4-dihydroxycinnamic acid(peak 5),chlorogenic acid(peak 4),3,4-dicaff eoylquinic acid(peak 16),3,5-dicaff eoylquinic acid(peak 17),4,5-dicaff eoylquinic acid(peak 18),luteolin(peak 20),quercetin(peak 19),5-caff eioylquinic acid(peak 1)and 4-caff eioylquinic acid(peak 9)in Lonicera japonica leaves may be the main active components of antioxidants.
