基于Au NP@CuS HNP复合材料的夹心型电化学免疫传感器及其在CEA检测中的应用

摘要 基于负载金纳米粒子(Au NP)的CuS中空纳米多面体(CuS HNP)复合材料(Au NP@CuS HNP)构建了一种夹心型电化学免疫传感器,用于癌胚抗原(CEA)的灵敏检测。CuS HNP外层和壳内空腔存在丰富的反应位点,具有高的催化活性;Au NP的负载提高了Au NP@CuS HNP整体的导电性和生物相容性。Au NP@CuS HNP用作信号标签,连接标记抗体(Ab)的同时能够高效催化HO还原产生电流信号。以金三角纳米板(Au TNP)作为基底材料,保证了电极界面的稳定性并且促进界面电子的转移。构建的电化学免疫传感器在1 pg/mL~50 ng/mL的CEA质量浓度范围内具有良好的检测能力,检出限(LOD)为0.37 pg/mL,为临床实现CEA的快速检测提供了一种新的方法。 A sandwich-type electrochemical immunosensor was constructed based on composite of CuS hollow nano polyhedrons(CuS HNPs) loaded with Au nanoparticles(Au NPs)(Au NPs@CuS HNPs) for the sensitive detection of carcinoembryonic antigen(CEA). CuS HNPs have abundant reaction sites in the outer layer and inner shell cavity, which have high catalytic activity. The loading of Au NPs improves the overall conductivity and biocompatibility of Au NPs@CuS HNPs. Au NPs@CuS HNPs are used as the signal labels to conjugate with secondary antibodies(Ab), meanwhile, can efficiently catalyze the reduction of HOto generate current signals. Au triangular nanoplates(Au TNPs) are used as substrate materials to ensure the stability of the electrode interface and promote the transfer of interfacial electrons. The prepared electrochemical immunosensor has good detection ability in the CEA mass concentration range of 1 pg/mL-50 ng/mL, and the limit of detection(LOD) is 0.37 pg/mL, which provides a new method for the rapid detection of CEA in clinical.
