摘要
目的:选择乳腺癌细胞作为研究对象,分析circPUM1对乳腺癌细胞的增殖、迁移及侵袭的影响及作用机制。方法:将MCF-7细胞分为MCF-7组、空载体组、circPUM1干扰组;circPUM1干扰组使用circPUM1 siRNA转染干扰circPUM1表达,空载体组使用空载体转染,MCF-7组不做其他处理;检测细胞增殖、迁移、侵袭能力及miR218-5p/IGF1水平。结果:MCF-7组、空载体组和circPUM1干扰组3组细胞增殖、迁移、侵袭活性存在明显差异,其中circPUM1干扰组细胞增殖、侵袭活性最低,迁移距离最小,且差异有统计学意义(P<0.05);MCF-7组、空载体组和circPUM1干扰组3组细胞miR218-5p、IGF1水平存在明显差异,其中circPUM1干扰组细胞miR218-5p水平最高、IGF1水平最低,且差异有统计学意义(P<0.05)。结论:circPUM1可能通过miR218-5p/IGF1调节乳腺癌细胞的增殖、迁移及侵袭活性。
Objective:To select breast cancer cells as the research object,and analyze the effects and mechanism of circPUM1 on the proliferation,migration and invasion of breast cancer cells.Methods:MCF-7 cells were divided into MCF-7 group,empty vector group,circPUM1 interference group;circPUM1 interference group was transfected with circPUM1 siRNA to interfere with circPUM1 expression,empty vector group was transfected with empty vector,MCF-7 group was not treated with other treatments;Detect cell proliferation,migration,invasion ability,miR218-5p/IGF1 level for detection.Results:The cell proliferation,migration and invasion activities of the MCF-7 group,the empty vector group and the circPUM1 interference group were significantly different.Among them,the circPUM1 interference group had the lowest cell proliferation and invasion activity,minimum migration distance,and the difference was statistically significant(P<0.05);MCF-7 group,empty vector group and circPUM1 interference group have significant differences in the levels of miR218-5p and IGF1.Among them,the circPUM1 interference group has the highest miR218-5p level and the lowest IGF1 level,and the difference is statistically significant(P<0.05).Conclusion:circPUM1 can regulate the proliferation,migration and invasion activities of breast cancer cells through miR218-5p/IGF1.
