摘要
目的 分析芪参还五胶囊对大鼠帕金森病相关基因核受体相关因子1(Nurr1)表达的影响及作用机制。方法 将30只SD大鼠均分芪参还五胶囊低、中、高剂量组及模型对照组和正常对照组,前4组大鼠于内侧前脑束单位注射6-羟基多巴胺建立帕金森病模型、对照组大鼠手术但不造模;造模后的第2天低、中、高剂量组分别大鼠给予0.93、1.87及3.75 g/(kg·d)剂量的芪参还五胶囊溶液灌胃、模型对照组和正常对照组给予等量生理盐水灌胃,连续7 d;于造模成功后的第7天及第14天时,比较各组大鼠黑质纹状体的谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽-S转移酶(GSH-ST)、白细胞介素-4(IL-4)、转化生长因子(TGF-β)及Nurr1 mRNA水平,检测黑质纹状体细胞的细胞活力、侵袭能力、凋亡率,比较各组大鼠黑质纹状体的神经突触数、突触后致密物(PSD)含量、活性区长度及磷酸化蛋白激酶B(p-AKt)、蛋白激酶B(AKt)蛋白表达水平。结果 造模后第7天及第14天时,与正常对照组相比,芪参还五胶囊低、中、高剂量组及模型对照组大鼠的黑质纹状体GSH、GSH-Px、GSH-ST含量,细胞活力、侵袭能力、突触个数、活性区长度、PSD含量、p-AKt蛋白表达水平均降低;IL-4、TGF-β、Nurrl mRNA水平,细胞凋亡率和AKt蛋白表达水平均升高,差异有统计学意义(P<0.05);与高剂量组相比,低剂量和中剂量组、模型对照组大鼠的黑质纹状体GSH、GSH-Px、GSH-ST含量、细胞活力、侵袭能力、突触个数、活性区长度、PSD含量、p-AKt蛋白表达水平均降低,IL-4、TGF-β、Nurr1 mRNA水平、细胞凋亡率、AKt蛋白表达水平均升高,且造模后第14天时低、中、高剂量组大鼠的黑质纹状体GSH、GSH-Px、GSH-ST含量、细胞活力、细胞侵袭能力、突触个数、活性区长度、PSD含量、p-AKt蛋白表达水平随着芪参还五胶囊剂量的升高而升高,IL-4、TGF-β、Nurr1 mRNA水平、细
Objective To investigate the mechanism of action of Qishenhuanwu capsule on(nuclearreceptor relatedfactor 1,Nurr1)gene expression in rats with Parkinson's disease(PD).Methods Thirty SD rats were equally divided into the low,middle,and high dose groups of Qishenhuanwu Capsule,model and normal groups.The first four groups were injected with 6-hydroxydopamine to establish rat with PD,and control group was given operation but without 6-hydroxydopamine.Low-,middle-and high-dose groups had been given respectively 0.93,1.87,and 3.75 g/(kg·d)of Qishenhuanwu capsule by gavage for 7 days since the 2^(nd) day after the successful establishment of the model,respectively,while model and normal groups had been daily given the same amount of normal saline by gavage.On the 7^(th) and 14^(th) day after successful modeling,glutathione(GSH),glutathione peroxidase(GSH Px),glutathione-S transferase(GSH-ST),interleukin-4(IL-4),transforming growth factor-β(TGF-β),and Nurr 1 mRNA in rat substantia nigra and striatum were compared between the groups.Cell viability,invasiveness and apoptosis rate of the cells in rat substantia nigra and striatum were detected.The number of synapses,the content of postsynaptic density(PSD),the length of active region and the expression levels of phosphorylated protein kinase B(p-AKT)and AKT in rat substantia nigra and striatum were compared between the groups.Results On the 7^(th) and 14^(th) day after modeling,the levels of GSH,GSH-Px,and GSH-ST,cell viability,invasiveness,number of synapses,length of active region,PSD,and p-AKt protein expression in rat substantia nigra and striatum in the low-,middle-,and high-dose groups as well as model group were decreased when compared with normal group,while IL-4,TGF-β,Nurr l mRNA,apoptosis rate,and AKt protein expression were significantly increased(P<0.05).When compared with high-dose group,the levels of GSH,GSH-Px,and GSH-ST,cell viability,invasiveness,number of synapses,length of active region,PSD,and p-AKt protein expression in rat substantia nigra and str
