羊口疮病毒青海株的分子鉴定及其遗传进化分析 被引量:2

摘要 青海祁连地区某规模化养殖场羊群疑似感染羊口疮,为准确鉴定此地羊口疮病原并对流行毒株进行遗传变异研究,采集疑似羊口疮病毒感染的羊唇部结痂组织并进行PCR鉴定,对鉴定的3株ORFV毒株的抗原基因B2L和F1L及毒力基因VIR和GIF分别进行克隆、测序及遗传进化分析。从病料中鉴定出3株ORFV,并分别命名为ORFV1/Ovis/QL/Qinghai/2021/China、ORFV2/Ovis/QL/Qinghai/2021/China和ORFV3/Ovis/QL/Qinghai/2021/China。经分析显示,ORFV1、ORFV2和ORFV3的B2L、F1L、VIR和GIF与数据库中ORFV参考株的相应的基因核苷酸序列和氨基酸序列同源性分别为99.21%~97.27%和99.74%~97.35%、99.12%~95.41%和99.12%~92.69%、98.19%~95.29%和96.72%~91.26%、99.62%~95.21%和100.00%~93.96%。基于ORFV的4个基因遗传进化树分析表明,中国青海祁连ORFV毒株分别聚在中国株的进化分支上(B2L基因),印度毒株和中国毒株的进化分支上(F1L基因),印度毒株、中国毒株和阿根廷毒株的进化分支上(VIR基因),中国毒株德国株、美国株、日本株、新西兰株、英国株、法国株、芬兰株的进化分支上(GIF基因);青海祁连ORFV流行株具有多样性,与疫苗株之间有一定的差异,流行株间也有差异。结果表明,该养殖场确实发生了羊口疮疫病,病原为ORFV,且ORFV来源可能不同,4种基因发生了较大的变异,有必要开展更为深入的流行病学调查,为防控青海地区的羊口疮疫病提供科学依据。 A large-scale farm in Qilian area of Qinghai Province was suspected of being infected with Orf.In order to accurately determine the pathogen of the suspected Orf disease and to study the genetic variation of the epi-demic strains of the Orf virus(ORFV).The scabs of infected with suspected ORFV were collected and identified by PCR molecular identification.The antigen gene B2L,F1L and the virulence gene VIR,GIF of the three identi-fied ORFV strains were cloned,sequenced and analyzed,respectively.The three ORFV strains were successfully i-dentified and named as ORFV1/Ovis/QL/Qinghai/2021/China,ORFV2/Ovis/QL/Qinghai/2021/China and OR-FV3/Ovis/QL/Qinghai/2021/China,respectively.The analysis results showed that the gene nucleotide sequence and amino acid sequence homology of the B2L,F1L,VIR and GIF of ORFV1,ORFV2 and ORFV3 were 99.21%~97.27%and 99.74%~97.35%,99.12%~95.41%and 99.12%~92.69%,98.19%~95.29%and 96.72%~91.26%,99.62%~95.21%and 100.00%~93.96%with the corresponding ORFV reference strains in the database,respectively.The genetic phylogenetic tree analysis based on the four genes of the three ORFV strains showed that the China Qinghai Qilian ORFV strains were clustered on the clade of the China strains(B2L);on the clade of the China strains and India strains(F1L);on the clade of the India strains,China strains and Argentina strains(VIR);on the clade of the China strains,Germany strains,USA strains,Japan strains,New Zealand strains,UK strains,France strains and Finland strains(GIF).The Qinghai Qilian ORFV epidemic strains were diverse,and with differences among them and the vaccine strains,and also with differences between the epi-demic strains.The results showed that the ORFV was identified as the pathogenic agent that causes the suspected cases of Orf in large-scale sheep farms in Qilian area of Qinghai,and the ORFVs in Qinghai Province might be from different sources,the four genes of ORFV were with mutation.It was necessary to carry out more epidemiologi-cal studies,and the present study will provide scientific
