益气养阴散结方对A549裸鼠肿瘤生长、瘤内脂肪沉积及血清瘦素、脂联素的影响

摘要 目的:观察益气养阴散结方对A549荷瘤裸鼠肿瘤生长、瘤内脂肪沉积及血清瘦素、脂联素的影响。方法:取30只BALB/c裸鼠,皮下接种A549细胞株复制荷瘤模型,随机分为5组,每组6只,即:模型组,顺铂组,益气养阴散结方低、中、高剂量组,模型组给予等量生理盐水灌胃,顺铂组给予顺铂4 mg/kg周2次腹腔注射,益气养阴散结方低中高剂量组予5、10、20 g/kg益气养阴散结方灌胃,连续给药8周。测量瘤径,绘制肿瘤生长曲线。末次给药次日取材,称瘤重,计算抑瘤率;进行油红O染色,观察瘤内脂肪沉积,ELISA检测血清瘦素、脂联素含量。结果:与模型组比较,中药高剂量组肿瘤体积显著减小,差异具有统计学意义(P<0.05);中药各剂量组平均瘤重显著降低,差异具有统计学意义(P<0.01或P<0.05),顺铂组、中药高剂量组抑瘤率接近。与模型组比较,中药各剂量组瘤内脂肪沉积显著减少,中、高剂量组瘦素显著降低,脂联素显著升高,脂联素/瘦素比值显著升高,差异均具有统计学意义(P<0.01或P<0.05)。结论:益气养阴散结方可显著抑制肿瘤生长,减小瘤内脂肪沉积,降低血清瘦素水平,提高血清脂联素水平,提高脂联素/瘦素比值。 Objective:To explore effects of Yiqi Yangyin Sanjie Decoction on xenograft growth,fat deposition in xenografts and serum leptin,adiponectin levels and adiponectin/leptin in A549 tumor-bearing nude mice.Methods:A total of 30 male BALB/c mice aged from 4 to 6 weeks were subcutaneously inoculated with A549 human lung cancer cell line via axilla to establish tumor-bearing nude mice models,and randomly divided into five groups:model group,Cisplatin group,Yiqi Yangyin Sanjie(YQYYSJ)Decoction low,medium,high dose groups,then,model group was given the same volume saline,Cisplatin group was intraperitoneal injected Cisplatin 4 mg/kg twice a week,YQYYSJ Decoction low,medium,high dose groups were given YQYYSJ Decoction 5,10,and 20 g/(kg·d)for 8 weeks.For exploring xenograft growth,diameters and weights of xenografts were taken,growth curves were plotted,tumor-inhibition rates were calculated.Oil red O staining was performed.Serum leptin,adiponectin levels and adiponectin/leptin ratios were measured.Results:Compared with model group,xenograft diameters of YQYYSJ Decoction high dose group decreased significantly(P<0.05).Compared with model group,xenograft weights of YQYYSJ Decoction low,medium,high dose groups decreased significantly(P<0.01 or P<0.05).Tumor-inhibition rates of Cisplatin and YQYYSJ Decoction high dose groups were close.Compared with model group,serum leptin level of YQYYSJ Decoction medium and high dose groups decreased significantly(P<0.01 or P<0.05),fat deposition in xenografts of YQYYSJ Decoction low,medium,high dose groups decreased,serum adiponectin level of YQYYSJ Decoction medium,high dose groups increased significantly(P<0.01),adiponectin/leptin ratios of YQYYSJ Decoction medium,high dose groups increased significantly(P<0.01 or P<0.05).Conclusion:YQYYSJ Decoction can significantly inhibited tumor growth,decreased fat deposition and serum leptin level,increased serum adiponectin level and adiponectin/leptin ratio.
