摘要
目的通过建立兔VX2乳腺癌模型,探讨高强度聚焦超声(HIFU)消融不同范围肿瘤后的转归及免疫效应。材料与方法采用包埋法建立兔VX2乳腺癌模型35只(对照组5只,3个消融组各10只),消融组在超声监控下分别行HIFU不同范围(50%、80%、100%)消融,比较超声与解剖学测量的消融体积,并通过组织切片染色及血浆ELISA检测比较各组消融后肿瘤细胞损伤、凋亡与增殖表达及血浆细胞因子(IL-2R、TGF-β1)变化情况。结果50%、80%、100%消融组通过超声及解剖后测得的肿瘤消融率结果一致(F=2.46、1.00、1.28,P均>0.05);组织切片显示80%消融组肿瘤细胞损伤明显,且可引起靶区交界处的肿瘤细胞凋亡,抑制细胞增殖效果明显;ELISA显示较50%消融组及对照组,80%及100%消融组在消融后血浆中免疫抑制因子IL-2R及TGF-β1表达水平显著降低,两组差异无统计学意义(P>0.05),且能明显抑制肿瘤生长,延长瘤兔生存期。结论当HIFU消融兔VX2乳腺癌的范围达80%及以上时,能显著抑制肿瘤增殖及生长,且能激活机体免疫反应,延长生存时间。
Purpose To investigate the results and immune effects of high-intensity focused ultrasound(HIFU)ablation of tumors of different sizes by establishing a rabbit VX2 breast cancer model.Materials and Methods The rabbit VX2 breast cancer model was established by embedding method(Thirty-five rabbits,5 in control group,10 in each ablation group),and HIFU ablation was performed in different ranges(50%,80%,100%)under ultrasound monitoring.The differences of ablation volume measured by ultrasound and dissection were compared.The tissue section staining and plasma ELISA were used to compare the tumor cell damage,apoptosis and proliferation expression,and changes in plasma cytokines(IL-2R,TGF-β1)after ablation in each group.Results The tumor ablation rate measured by ultrasound and dissection of each ablation group(50%,80%,100%)were consistent(F=2.46,1.00,1.28,all P>0.05);HE staining showed that the 80%ablation group had obvious damage to tumor cells,and could cause tumor cells at the junction of target area to atrophy,and had anobvious effect on inhibition of cell proliferation.ELISA results showed that compared with the 50%ablation group and control group,the 80%and 100%ablation groups had significantly lower expression levels of IL-2R and TGF-β1 after ablation,there was no statistical difference between the them(P>0.05),and they can significantly inhibit tumor growth and prolong survival of tumor rabbits.Conclusion When the range of HIFU ablation of rabbit VX2 breast cancer reaches 80%or more,it can significantly inhibit tumor proliferation and growth,and can activate the body's immune response and prolong survival time.
