摘要
为了解伊犁马携带病毒的类型,本研究利用高通量测序技术检测伊犁昭苏县142份伊犁马马驹鼻拭子样品。病毒组学分析显示,伊犁马马驹鼻拭子样品携带人巨细胞病毒(Human cytomegalovirus,HCMV),根据注释到的HCMV UL126基因序列,设计特异性引物,对样品进行PCR检测。结果显示,伊犁马马驹鼻拭子样品HCMV阳性率为7%(10/142),进一步调查显示伊犁马流产胎儿肺组织样品中该病毒阳性率为57%(53/93),表明该病毒感染伊犁马。另外,研究表明马场工作人员HCMV阳性率为37.5%(15/40),且大部呈HCMV阳性马驹和流产的妊娠母马都和HCMV阳性工作人员有密切接触史,提示马源HCMV可能来源人。核苷酸同源性分析显示本研究鉴定的马源和人源HCMV UL126基因的核苷酸序列同源性为99.6%~99.8%,与国外人源HCMV UL126基因的核苷酸序列同源性为97.8%~100%。遗传进化树分析显示本研究鉴定的HCMV与美国人源HCMV亲缘关系较近。本研究首次鉴定马源HCMV,为其致伊犁马流产防控和动物源HCMV流行病学调查奠定基础。
Viral metagenomic sequencing was applied to 142 nasal swabs collected from Yili horses in Zhaosu(China)to understand the types of viruses carried by these animals.Metagenomic data showed that the human cytomegalovirus(HCMV)circulated in samples.According to the HCMV reference strain Toledo/Towne Chimera 4,specific primers against the UL126 gene were designed to verify the results of viral metagenomics.Polymerase chain reactions showed that 10 nasal swabs of Yili horses were positive for the HCMV(7%,10/142),The prevalence of viruses in aborted Yili horses was 57%(53/93).Sequencing indicated that the nucleotides among UL126 of the HCMV shared 99.6%–99.8%homology,and shared 97.8%–100%nucleotide-sequence identity,with the UL126 genes of human-origin HCMV reference strains.A phylogenetic tree based on the sequences of the UL126 gene showed that the horse-origin and human-origin HCMV genes we identified were clustered in an independence lineage.These results provide new evidence that the HCMV can infect Yili horses in China,and may be a causative agent for abortions in Yili horses.
