摘要
目的探讨利拉鲁肽对糖皮质激素性骨质疏松大鼠骨代谢指标的影响。方法选取12周龄健康雌性Wistar大鼠80只,体质量200~240 g,采用随机数字表法分为模型组、阿仑膦酸钠组、利拉鲁肽组和对照组,每组20只。模型组、阿仑膦酸钠组、利拉鲁肽组肌肉注射地塞米松2 mg/kg,每周2次,共注射12周,建立糖皮质激素性骨质疏松模型,对照组注射同等剂量0.9%氯化钠注射液。造模成功后,利拉鲁肽组皮下注射利拉鲁肽200μg/(kg·d),对照组和模型组皮下注射同等剂量0.9%氯化钠注射液,阿仑膦酸钠组给予阿仑膦酸钠0.16 mg/(kg·d)研磨后溶于2 ml 0.9%氯化钠注射液灌胃。采用酶联免疫吸附测定法检测4组外周血清骨代谢指标[骨钙素(BGP)、骨特异性碱性磷酸酶(BALP)、Ⅰ型前胶原N-端前肽(PⅠNP)、Ⅰ型胶原交联C-末端肽(CTX-Ⅰ)],采用双能X线骨密度仪检测大鼠腰椎、股骨骨密度,采用电子万能试验机测定右侧股骨生物力学指标(最大弯曲荷载、弹性模量),采用Micro-CT扫描大鼠左侧股骨,并定量分析骨小梁厚度(Tb.Th)、骨小梁数量(Tb.N)、骨小梁连接密度(Conn.D)、骨小梁体积(BV/TV)比率。结果模型组腰椎及股骨骨密度均小于对照组、阿仑膦酸钠组及利拉鲁肽组,差异有统计学意义(P<0.05);阿仑膦酸钠组、利拉鲁肽组与对照组腰椎及股骨骨密度比较差异均无统计学意义。模型组最大弯曲荷载和弹性模量均低于对照组、阿仑膦酸钠组、利拉鲁肽组,差异有统计学意义(P<0.05);阿仑膦酸钠组、利拉鲁肽组与对照组最大弯曲荷载及弹性模量比较差异无统计学意义。模型组左侧股骨Tb.Th薄于对照组、阿仑膦酸钠组、利拉鲁肽组,Tb.N少于对照组、阿仑膦酸钠组、利拉鲁肽组,Conn.D小于对照组、阿仑膦酸钠组、利拉鲁肽组,BV/TV低于对照组、阿仑膦酸钠组、利拉鲁肽组,差异有统计学意义(P<0.05);阿仑膦酸钠组、利拉鲁
Objective To investigate the effects of liraglutide on bone metabolism indexes in rats with glucocorticoid-induced osteoporosis.Methods 8012-weeks-old healthy femaleWistar rats with body weight of 200-240 g were selected,and they were divided into model group,alendronate sodium group,liraglutide group and control group according to random number table method,with 20 rats in each group.The model group,alendronate sodium group,liraglutide group were injected intramuscularly with dexamethasone 2 mg/kg,twice a week,for a total of 12 weeks,to establish a glucocorticoid-induced osteoporosis model,and the control group was injected with the same dose of 0.9%sodium chloride injection.After successful modeling,the liraglutide group was subcutaneously injected with liraglutide 200μg/(kg·d),the control and model group were subcutaneously injected with the same dose of 0.9%sodium chloride injected,and the alendronate sodium group were given alendronate sodium 0.16 mg/(kg·d)after grinding and then dissolved in 2 ml of 0.9%sodium chloride injection by gavage.The peripheral serum bone metabolism indexes(bone gla protein[BGP],bone alkaline phosphatase[BALP],procollagen typeⅠN-terminal propeptide[PⅠNP],C-terminal telopeptide of typeⅠcollagen[CTX-Ⅰ])were detected by enzyme-linked immunosorbent assay among the four groups,the bone mineral density of the lumbar spine and femur in rats were detected by Dual-energy X-ray absorptiometry,the biomechanical indexes of right femur(maximum bending load,elastic modulus)were detected by electronic universal testing machine,the microstructure of rat left femur were scaned by Micro-CT,and the trabecular thickness(Tb.Th),trabecular number(TB.N),trabecular connection density(Conn.D)and bone volume/tissue volume ratio(BV/TV)were measured.Results The bone mineral density of the lumbar spine and femur in the model group was lower than that in the control group,the alendronate sodium group and the liraglutide group,and the difference was statistically significant(P<0.05);there was no signi
