摘要
采用超声波辅助法提取金丝皇菊总黄酮。在单因素试验的基础上,采用响应面法优化金丝皇菊总黄酮的提取工艺条件,并通过体外抗氧化试验对其抗氧化能力进行探究。结果表明:金丝皇菊总黄酮最优提取工艺参数为超声功率240 W、乙醇体积分数81%、提取时间42 min,在此条件下总黄酮得率为64.92 mg/g,金丝皇菊总黄酮提取物对DPPH·、·OH、·O_(2)^(-)和ABTS^(+)·均有显著的清除作用,且抗氧化活性与其质量浓度存在良好的量效关系。当金丝皇菊总黄酮质量浓度为0.10 mg/mL时,对DPPH·、·OH、·O_(2)^(-)和ABTS^(+)·的清除率分别为64.36%、89.42%、78.99%和63.90%,IC_(50)分别为0.058、0.036、0.043和0.043 mg/mL。
The total flavonoids from Dendranthema morifolium were extracted by ultrasonic-assisted method.On the basis of single factor experiment,the extraction conditions of total flavonoids from Dendranthema morifolium were optimized by response surface method,and its antioxidant capacity was studied by antioxidant test in vitro.The results showed that the optimum extraction parameters of total flavonoids from Dendranthema morifolium were as follows:ultrasonic power 240 W,volume fraction of ethanol 81 %,extraction time 42 min.Under these conditions,the yield of total flavonoids was 64.92 mg/g.The total flavonoids extract from Dendranthema morifolium had significant scavenging effects on DPPH;·O_(2)^(-)H,·O_(2)^(-)and ABTS^(+)·,and there was a good dose-effect relationship between antioxidant activity and its mass concentration.When the total concentration of total flavonoids in Dendranthema morifolium was 0.10 mg/mL,the scavenging rates of DPPH·,·O_(2)^(-)H,·O_(2)^(-)and ABTS^(+)· were 64.36 %,89.42 %,78.99 % and 63.90 %,respectively,and the IC_(50)was 0.058,0.036,0.043 and 0.043 mg/mL,respectively.
