摘要
目的建立不同产地不同采收期金银花中挥发性成分的GC-MS指纹图谱并结合化学计量学手段进行成分分析。方法金银花挥发油的分析采用TG-5MS毛细管柱(30 m×0.25 mm,0.25μm);进样口温度:280℃;载气:氦气;分流比:10∶1;离子源温度:300℃;传输线温度:300℃;四级杆温度:150℃。采用GC-MS分析不同产地不同采收期的金银花挥发性成分,建立9批样品的指纹图谱,对其进行相似度评价,确定共有成分,并利用主成分分析(PCA)和热图分析比较样品质量。结果9批样品GC-MS指纹图谱有18个共有峰,相似度均大于0.9,挥发性成分种类相似,但相对含量仍存在差异。结论建立的方法精密度、重复性和稳定性良好,可用于金银花质量的控制。
Objective To establish a method based on GC-MS combined with fingerprint and chemometrics for the study on compositional changes of volatile oils of honeysuckle at different harvesting periods in different origins.Methods The analysis of honeysuckle volatile oil was performed on a TG-5 MS capillary column(30 m×0.25 mm, 0.25 μm).Inlet temperature, ion source temperature and transmission line temperature were 280 ℃,300 ℃ and 300 ℃,in that order.And the split ratio was 10∶1.The volatile components of honeysuckle from different origins and different harvesting periods were analyzed by GC-MS.Then, the fingerprint profiles of the nine batches of samples were established, similarity was evaluated and shared peaks were identified.Meanwhile, The chemical compo sitions were compared by principal component analysis( PCA) and heat map analysis.Results The fingerprints of the nine batches of samples had 18 common peaks with similarities greater than 0.9.The volatile components were similar in type, but there were still differences in relative contents.Conclusion The established method has good precision, reproducibility and stability and can be used for the quality control of honeysuckle.
