RFRP-3对雌性仔猪生长性能、血清生化指标及葡萄糖代谢的影响

摘要 为了探讨RF-酰胺相关肽-3(RFRP-3)对雌性仔猪生长性能、血清生化指标及葡萄糖代谢的影响,选取6周龄雌性仔猪15头,随机均分为3组,分别为对照组、RFRP-3低剂量组和高剂量组,每天分别腹腔注射0.9%生理盐水、0.1 mg/mL RFRP-3和1 mg/mL RFRP-3两次,每次1 mL,连续14 d。结果:与对照组相比,腹腔注射RFRP-3极显著增加雌性仔猪平均日增重(P<0.01),显著降低料重比(P<0.05);肥胖指标POI指数极显著增加(P<0.01);白色脂肪和胰腺比重显著增加(P<0.05,P<0.01);血清生化指标分析发现,腹腔注射RFRP-3仔猪血清中甘油三酯、葡萄糖含量极显著升高(P<0.01),高剂量组肝脏质量和低密度脂蛋白含量显著升高(P<0.05);空腹血糖试验发现RFRP-3极显著诱导血糖总量升高(P<0.01);肝脏PEPCK mRNA表达水平显著上升(P<0.05),GCK mRNA表达水平极显著下降(P<0.01)。本试验表明,腹腔注射RFRP-3可提高雌性仔猪生长性能并伴随血糖和血脂浓度的升高,且这种生理功能的发挥与肝脏糖代谢相关因子的变化密切相关。 This study was to investigate the effects of RF amide-related peptide 3(RFRP-3)on the growth performance, serum biochemical indexes and glucose metabolism in piglets. 15 female piglets at 6 weeks of age were selected and randomly divided into three groups: the control group(administered with 0.9% physiological saline), the RFRP-3 low dose group(with 0.1 mg/1 mL RFRP-3) and the high dose group(with 1 mg/1 mL RFRP-3), by intraperitoneal injection of 1 mL of the prepared regents twice a day for 14 consecutive days. The results showed that, compared with the control group, intraperitoneal injection of RFRP-3 extremely significantly increased the mean daily weight gain of the female piglets(P<0.01) and significantly reduced their feed to weight ratio(P<0.05). Among the obesity indicators, the POI index increased extremely significantly(P<0.01), the white fat and pancreatic masses were significantly or extremely significantly increased(P<0.05;P<0.01). In the serum biochemical analysis, the triglyceride and glucose levels were found to be highly significant(P<0.01), and the liver mass and the LDL level were found to be significantly increased in the high-dose RFRP-3 group(P<0.05). RFRP-3 was found to have induced a highly significant increase in the total blood glucose in the fasting glucose test(P<0.01);and the liver PEPCK mRNA expression levels of the piglets were significantly increased(P<0.05), but their GCK mRNA expression levels were highly significantly decreased(P<0.01). In conclusion, intraperitoneal injection of RFRP-3 improved the growth performance of the female piglets, shown by increased blood glucose and lipid concentrations, and these physiological functions were closely related with the changes in the factors involving liver glucose metabolism.
